Purpose:This experiment aimed at Yukui square(YKR)of three nitrobenzene sulfonic acid(TNBS)induced ulcerative colitis rat colonic mucosal barrier function in the role of regulation,based on the tight junction protein Claudin-2 and Claudin-7 and immune protein MBL and MASP-2 protein expression studies,explore Yukui in intestinal mucosal ulcer repair function colitis rats and the regulating effect on the intestinal immune function.Material and method:60 adult male SD rats were randomly assigned to 6 groups: A.blank group,B.model group,C.YKRH dose group,D.YKRI dose group,E.YKRL dose group,F.mesalazine group.Except the normal control group,B,C,D,E,F group rats were induced by TNBS disease UC SD rats before modeling fasting 24 h,touch the back of rats before enema stimulation distal emptying stool,by intraperitoneal injection of chloral hydrate anesthesia,a polyethylene 0.2cm diameter pipe 8cm into the colon,slowly poured into the TNBS solution(TNBS 5% aqueous solution,and the preparation of anhydrous ethanol,1:1ethanol concentration of 50% by volume),the dose of 60mg/kg,a continuous 1min upside down after enema,modeling second days showed that all rats appeared watery and bloody stool,began intragastric administration of traditional Chinese medicine and the beauty of salad,A,B group rats were given 0.5% sodium carboxymethyl cellulose,the dosage according to the human body conversion formula to convert [22]C,D,E group rats were given different doses of Chinese herbal medicine,Chinese medicine high dose group14mg/g/d,normal dose group 7 Mg/g/d,low dose of Chinese medicine group 4mg/g/d dose,F group rats were given 0.2g/kg/d the beauty of salad suspension was dissolved in 5ml distilled water given orally administered continuously for 14 days,the rats were sacrificed,wasassessed rats DAI,CDMI,TDI,and colon were used immunohistochemical technique to detect the expression of Claudin-2 and Claudin-7 and MBL and MASP-2 protein were analyzed by statistical software.Results:1.YKR can effectively improve the symptoms of UC rats,improve the weight of rats,fecal occult blood,in effect,high dose of YKR group > beauty group >YKR salad diazoxide dose group >YKR low dose group.TNBS induced UC rats in the use of YKR high dose of Chinese medicine,the rats DAI,CMDI and TDI score,the effect was not statistically significant with the mesalazine group,all effectively reduce the score.YKR low dose group,rats DAI,CMDI,TDI score and model group no difference,so no effect;2.TNBS induced colonic mucosa damage in SD rats,increased Claudin-2,decreased Claudin-7,influenced intestinal mucosal remodeling and increased permeability.In the use of YKR,high dose group can reduce Claudin-2,increase Claudin-7,effectively improve intestinal mucosa recovery,low dose group is not obvious;3.TNBS induced SD rats colon can make the immune protein MBL protein and MASP-2protein decline.TNBS induced SD rats in the use of YKR,high dose group can make MBL protein and MASP-2 protein recovery,low dose group is not obvious.Conclusion:1.high school YKR can effectively improve the pathological status of UC rats,low YKR effect is lower.2.high school YKR can adjust the normal restoration of Claudin-2 and Claudin-7,effectively restore intestinal mucosal function,low dose ineffective.3.YKR high dose group can effectively adjust intestinal immune function,and promote the normal development of MBL and MASP-2,low dose ineffective.
