This research include two parts,the first part is the research of detection methods about tumor markers. In the basis of traditional method, Developed a slides protein microarray for detection tumor markers, and established the method of bacteria fluorescent enzyme bioluminescent immune analysis. The second part is research about Medicinal plants Rumex japonicus Houtt quality ecology, the main research of medicinal plants Rumex japonicus Houtt growth rule under the different habitat and the effective composition activity, and studied in different habitat Rumex japonicus Houtt of inner genetic diversity changes.Processed the slides with acetone, and modified it with agarose membrane. Pointed monoclonal antibody of anti-tumor markers by manual point. After preparation the slides protein microarray, detection the AFP in serum by this protein chip, meanwhile, compared with NC chips kit. Completed the research of slides protein microarray for tumor markers.Preparation G6PDH-AFP by SPDP, determine the optimum conditions of antigen-antibody reaction and conditions of luminous system, established the bioluminescent immune analysis method. Using the bioluminescent immune analysis to detect the serum AFP. Meanwhile, compared with ELISA method, and better than it.Research the quality ecology of medicinal plants Rumex japonicus Houtt in Jilin City. Study the main chemical compositions by HPLC. and study antifungal activity of mainly chemical composition by FCM, and study the Genetic diversity by ISSR. Born plants for two or three years the anthraquinone content high, the best harvest time is 7 months, anthraquinone have very good antifungal activity.The results of ISSR analysis indicate that among populations.the percentage of polymorphic loci(P)is from 25.93% to 37.04%. the gene diversity(H) is 0.1751 and Shannon Index(1)is from 0.2634. Analysis of genetic differentiation within species, the results showed that the genetic differentiation coefficient (Gst) among populations was 0.3662,demonstrated that the among-population component accounted for 36.62% of the total variation, while the within-population component accounted for 63.28% of the total variation, while the within-population component accounted for 63.28%. The gene flow among populations was 0.8654. Analysis of genetic differentiation among populations and these four populations could be clustered into one groups using unweighted pair group method with arithmetic mean (UPGMA)..there are genetic diversity exists within populations.and the genetic diversity is positive correlation with the ability to adapt to the environment of the Rumex japonicus; Genetic differentiation existed among the four populations, but the genetic structure change is not big.Based on the above results, in different habitat the quality of medicinal plants Rumex japonicus Houtt without change.
