Objective:The purpose of this research is to observe the preventive and therapeutic effects of Qinganyixian Decoction and its disconnected prescription on rat hepatic fibrosis, and discuss its mechanism of action.Methods:110 Wistar rats were divided into 9 groups according to the principle of randomization. The control group was given normal diet, free drinking water, The remaining groups were given subcutaneous injection with 40% CCl4-oil solution on their limbs or back twice a week in accordance with 0.3 ml/100g, and they were lavaged with the herbs or water for 8 weeks.8 weeks later, after a success model checking, kill the remaining rats. Measure the rat body weight and liver wet weight, alculate the liver organ index. Detect the liver function and serum hepatic fibrosis markers (HA, LN, C-IV).Detect rat liver tissue homogenate MDA content and SOD activity, assay the concentration of TNF-α, TGF-β1, MMP-2 and TIMP-2 of the rat liver tissue by means of ELISA, detect the expression of TGF-β1mRNA using RT-PCR, test the apoptosis of HSC with TUNEL-step detection. Liver histopathology observation include of observing the liver tissue inflammatory degree and collagen deposition grade using HE and Masson staining; observing the expression of a-SMA in the rats liver by means of immunohistochemical method and quantitatively analyzing the results.Results:After Statistical analysis experimental data turned up that the Qinganyixian decoction and its disconnected prescription can decrease the serum ALT, AST, MDA content, increase ALB and SOD activity, reduce the degree of liver inflammation and and the fat quality peroxidation significantly. The effect of the whole description was better than the disconnected prescriptions.Qingrejiedu herbs and Liangxuehuoxue herbs play the most important role. The Qinganyixian decoction and its disconnected prescription can decrease the expression of a-SMA positive cells of liver tissue significantly, inhibit the activation of stellate cells. The apoptosis rate of activated hepatic stellate cells in the therapeutical groups increased, but there was no significant difference compared with the model group. TGF-β1 from the mRNA to protein level expressed abnormal significantly, increasing with the severity of liver fibrosis. The Qinganyixian decoction and its disconnected prescription can reduce its expression significantly. There was a significant difference (P<0.01, P<0.01) compared with model group. The Qinganyixian decoction and its disconnected prescription may significantly reduce the TNF-αlevel in liver tissue, especially the Qingrejiedu herbs. The Qinganyixian decoction and its disconnected prescription can reduce TIMP-2, increase MMP-2/TIMP-2 ratio, the Liangxuehuoxue herbs plays the major role.Conclusions:Qingganyixian Decoction and its disconnected prescription can prevente and treate hepatic fibrosis, and its mechanisms are:1. Reduce liver cell degeneration, necrosis and lipid peroxidation levels, improve liver inflammation, improve antioxidant capacity, promotie liver cell repair, and thereby reduce or eliminate predisposing factors of the liver fiber in order to block the initial part of hepatic fibrosis.2. Iinhibit the HSC activation and promote apoptosis of activated HSC to reduce the number of muscle fibroblasts. Thus it can reduce ECM synthesis to prevent the process of hepatic fibrosis.3. Reduce the expression of promoting hepatic fibrosis factors:TGF-β1 and TNF-αin the liver, then it can block the network transmission of the cytokine in the formation of fibrosis.4. Reduce TIMP-2 concentration in the liver tissue, increase MMP-2/TIMP-2 ratio to accelerate the degradation of ECM formed.
