Purpose:To explore the role of mechanisms to improve sleep of SNSC. Method:the experiment begin with sodium pentobarbital collaborative experiment. The research about incubation period and sleep period in mice try to make it sure that the stady on improve sleep of NSEC.The research about dose-response relationship and time-concentration of SNSC on improve sleep.The compare bettween SNSC and SNS on the role of mechanisms. based on that, We carried out the mechanism research on improve sleep of SNSC, also based on GABAergic nervous system,serotonergic nervous system.NO signaling pathway, theory of sleep mediated by SP, IL-1βand TNF-α, PGE2,PGD2,IL-6,used a series of research methods and technology such as sodium pentobarbital collaborative experiment,enzyme linked immunosorbent assay,chemical colormetry.The results:Part one: The research about the role of SNSC on improve sleep1. The research's result showed that if continuous intragastric administration 7 days by SNSC( 33.00mg/ml,660.00mg/kg).they can extend the pentobarbital sodium-induced sleep time in mice and shorten the incubation period.which were significantly higher than that of control group ( p<0.01).2.The research's result about dose-response relationship on improve sleep of SNSS showed that the minimum effect dose is 330.00 mg/kg.it can shorten the incubation period of the model mice.which were significantly different than that of control group (p<0.05); the best dose is 660.00 mg/kg which register as extend pentobarbital sodium-induced sleep time in mice and shorten the incubation period. which were significantly different than that of control group (p<0.01);the efficacy will abate when the dose is 1320.00 mg/kg, but the register also extend pentobarbital sodium-induced sleep time in mice and shorten the incubation period.which were significantly different than that of control group (p<0.01).3.The research's result about time-concentration relationship on improve sleep of SNSC showed that the dose is 660.00mg/kg( i.p), it will show the efficacy on the fouth day, register as extend pentobarbital sodium-induced sleep time in mice.(p<0.05);and shorten the incubation period, (p<0.01);if continuous to give the SNSC it will enhance(p<0.01).4. The research's result about the different bettween the role of mechanisms to improve sleep of SNSC and SNS showed that when choose the best dose and the earliest take effort time.both of them can shorten the incubation period in mice. There is no significantly difference. But there was significantly difference on extend the pentobarbital sodium-induced sleep time in mice (p<0.01).Part two: the research of mechanisms to improve sleep of SNSC1. The research's result about the affect on the sleep in mice of pentobarbital with SNSC combined application of Flumazenil showed that it can extend the pentobarbital sodium-induced sleep time in mice.which were not significantly different than that of control group (P>0.05). There were significantly difference compare than the SNSC group (P<0.05).2. SNSC combined application of 5-HT can significantly prolong sodium pentobarbital-induced sleep time in mice.which were significantly different than that of control group (P<0.01).3. SNSC combined application of PCPA can prolong sodium pentobarbital-induced sleep time in mice.which were not significantly different than that of control group (P>0.05).4. SNSC combined application of L-Arg can prolong sodium pentobarbital-induced sleep time in mice.which were not significantly different than that of control group (P>0.05).5. SNSC combined application of L-NAME can prolong sodium pentobarbital-induced sleep time in mice.which were significantly different than that of control group (P>0.05).6.From the study on the influence on ON content of brain,thalamencephalon and brain stem showed that it can significantly increase the ON content of brain on the fouth day which continue togive the SNSC(i.p).which were significantly different than that of control group (P<0.05); it can significantly increase the ON content of thalamencephalon and brain stem.which were significantly different than that of control group (P<0.01).7.From the study on the influence on SP content of brain,thalamencephalon and brain stem on the fouth day which continue to give the SNSC(i.p) showed that there were not significantly different than that of control group (P>0.05).8.From the study on the influence on IL-1βcontent of brain,thalamencephalon and brain stem showed that it can significantly reduce the IL-1βcontent of brain on the fouth day which continue to give the SNSC(i.p). which were significantly different than that of control group(P<0.05); There is a little increase in the IL-1βcontent of thalamencephalon and brain stem. But there is not significantly different compared with that of control group (P>0.05).9.From the study on the influence on TNF-αcontent of brain,thalamencephalon and brain stem showed that it significantly increase the TNF-αcontent of brain and brain stem on the fouth day which continue to give the SNSC(i.p). which were significantly different than that of control group (P<0.05); it can significantly reduce the TNF-αcontent of thalamencephalon.which were significantly different than that of control group (P<0.01).10.From the study on the influence on PGD2 content of brain,thalamencephalon and brain stem showed that it can significantly reduce the PGD2content of brain on the fouth day which continue to give the SNSC ( i.p).which were significantly different than that of control group (P<0.05); There is a little reduce in the PGD2 content of thalamencephalon and brain stem.which were not significantly different than that of control group (P>0.05).11.From the study on the influence on IL-6 content of brain,thalamencephalon and brain stem showed that it can significantly reduce the IL-6 content of brain on the fouth day which continue to give the SNSC ( i.p).which were significantly different than that of control group (P<0.05); it can significantly increase the IL-6 content of thalamencephalon .which were significantly different than that of control group (P<0.05); There were a little increase in the IL-6 content of brain stem.which were not significantly different than that of control group (P>0.05).12.From the study on the influence on PGE2 content of brain,thalamencephalon and brain stem on the fouth day which continue to give the SNSC(i.p)showed that there were not significantly different than that of control group (P>0.05).Summary:1,SNSC can improves sleep, the best dose is 660.00mg/kg by intrapertioneal injection,the earliest take effort time is the fouth day.2,GABA,5-HT,NO,IL-1β,TNF-α,PGD2,IL-6 participate in improveing sleep of SNSC.
