Object:Find out the Cirenshen’s effective part which improves sleep and explore its mechanism.Method:This experiment with ICR mice as the research object,firstly by pentobarbital sodium collaborative experiment, verified the Cirenshen decoction on improving sleep effect;on this basis, the Cirenshen decoction with macroporous adsorption resin separation into10%,30%,60%and three different concentrations of ethanol elution fraction,using amyl barbital sodium collaborative experiment further identification of Cirenshen on improving sleep effect effective part;on the basis of effective parts of Cirenshen on improving sleep effect dose effect relationship and time effect relation;finally use pentobarbital sodium collaborative experiment method of effective parts of Cirenshen on improving sleep effect correlation with GABA,and respectively were determined in mice hippocampus the cortex, hypothalamus, DA, GLU,NE,5-HT,NO,NOS and other substances,to carry out effective parts of Cirenshen on improving sleep effect mechanism research.Results:1.Cirenshen decoction on the threshold dose of pentobarbital sodium in mice induced by sleep time effects:The experimental results show that,compared with blank group,positive control group significantly shortened sleep latency of mice and significantly prolong sleep time (P<0.05),indicating that the test method is reliable.Compared with the blank control group,Cirenshen every dose group significantly prolonged pentobarbital sodium in mice induced by sleep duration (P<0.05);Cirenshen every dose group of sleep latency shortening trend appears, in which64g/kg group and the control group had significant difference (P<0.05).2.Cirenshen decoction on under threshold dose of pentobarbital sodium in mice induced by sleep incidence:The experimental results show that, the experimental results show that, compared with blank group, positive control group significantly shortened sleep latency of mice and significantly prolong sleep time(P<0.05),indicating that the test method is reliable.
