Reproduction and Development are important questions in the field of biological research. As the highly differential male germ cell, the genes of sperm have strict relation of time and space during the proceeding of spermatogenesis and maturation, especially the synthesis and transformation of sperm membrane proteins, the interaction of epididymal protein factors with sperm paly the important role in the spennatogenesis, capacitation, fusion of sperm and egg. So, it's very important to take research on the structure , function and gene expression of some membrane protein of sperm and epididymis.YWKII and BS-17 are two kinds of human sperm membrane protein. some research had been done on their structure and function. But the completely full length of their cDNAs have not been obtained, especially in the 5' region of cDNAs. Our research is focused on amplification of cDNAs HSD-2 encoding YWKII and BS-17 with 5' regions of unknown sequence by using simple, efficient and sensitive technique—RACE (Rapid Amplification of cDNA Ends). The fresh template mRNA is extracted from human testis. In order to isolate the 5' end of the cDNAs, a gene specific primer(GSP1) is used to perform reverse transcription. The specific first-strand cDNA is then purified and polyadenylated using terminal deoxynucleotidyl tranferase (TdT) and dATP. This synthetic poly(A) tract serves as the target for a poly(T)-containing anchor primer. These tailed-cDNAs are finally amplified by using oligo(dT)-anchor primer and another gene specific primer (GSP2) .When necessary, nested PCR can be performed. The amplified PCR products are cloned and sequenced.The complete cDNA of 1890bp of BS-17 was identified and sequenced, An open reading frame of 1632bp encodes 544 amino acid residues with a computed molecular mass of 60Kda.The complete cDNA of 3648bp of HSD-2 was identified and sequenced, An open reading frame of 2289bp encodes 763 amino acid residues with a computed molecular mass of 87Kda.
