Spermatogenesis is a series of highly ordered cell differentiation process, which involves the expression of many genes under precise temporal and spatial regulation. The cloning and identification of these differentially expressed genes is of great value in delineating the mechanism of spermatogenesis. Such a study can also provide the foundation and cues to investigate molecular basis of immunological infertility and to develop immunocontraceptives.Some ESTs isolated from rat seminiferous tubule using the method of DDRT-PCR combined with segmentation technique of seminiferous tubule or searched from GenBank were selected and used to screen human and rat testis cDNA libraries. Eight novel full-length cDNAs were isolated, identified, designated as HSD-4, RSD-4, RSD-5, Rat DNA J-like protein cDNA, tSH3pl3, RSD-6, RSD-7, HSD-3 and assigned the Accession Numbers : AF162680, AF315468, AF146738, AF154849, AF227439, AF271155, AF315467 and AF144487. In the present thesis, the cloning and characterization of these cDNAs are described.HSD-4 and RSD-4 were isolated from human and rat testis cDNA libraries. HSD-4 cDNA consists of 2497bp with an open reading frame (ORF) of 738bp, encoding a polypeptide of 245 amino acids. The gene was assigned the accession number AF162680. RSD-4 cDNA consists of 779bp, codes a polypeptide of 209 amino acids and assigned the accession number AF315468.
