The Study On The Compatibility Of Zhishi And Baizhu And The Mechanism Of Action Of Zhizhu Yin In Promoting Stomach Intestine Dynamia

Functional dyspepsia (FD) is a common disease of digestive system. Dyskinesis of stomach intestine is the main reason of functional dyspepsia (FD) in pathophysiology. Planning treatment according to diagnosis for FD have obvious ascendancy and prospect of development.Zhizhu Decoction which is chosen in the book named Synopsis of Golden Chamber has good curative effect on the treatment of FD. The objective of this experiment is to find the best proportion of the dosage in the compatibility of Zhishi and Baizhu and to approach the function mechanism of Zhizhu Yin by means of observing the effectiveness upon improving peristalsis of stomach intestine.Objectives: Research the compatibility of Zhishi and Baizhu and the function mechanism of Zhizhu Yin in promoting peristalsis of stomach intestine.Methods: Observe the effects that only using Zhishi or Baizhu and the compatibility of Zhishi and Baizhu have on the gastrointestinal motility of normal mouse. Observe the effects that only using Zhishi or Baizhu and the compatibility of Zhishi and Baizhu have on the containment in the stomach intestine of mouse induced by atrop. Observe the effects that only using Zhishi or Baizhu and the compatibility of the two drugs have on the motilin (MTL) substance P (SP) gastrin (GAS) vasoactive intestinal peptide (VIP) somatostatin (SS) and neurotensin (NT) in the FD modle rats. Approach the mechanism of action by means of observing the D and M endocrine cell, acetylcholine esterase (AchE) nitric oxide synthase (NOS) and gut hormone receptor (MTL-R1AmRNA and VIPR₁mRNA) in the stomach intestine of model rats.Results: All the three different proportions, that are 1: 1,1: 2 and 2: 1 in the compatibility of Zhishi and Baizhu, have the function to promote gastric emptying and intestine driving and this function is superior to that of using single drug. The proportion of 1:1 called Zhizhu Yin have the functions to increase MTL and decrease VIP. Zhizhu Yin can degrade the express of D endocrine cell in the sinus ventriculi and upgrade the express of M endocrine cell in the dodecadactylon and AchE and NOS in the sinus ventriculi janitor and dodecadactylon. It can upgrade the express of Ach and degrade the express of NO. It can up-regulate the express of MTL-R1AmRNA and downregulate that of VIPR₁mRNA.Conclusions: The three compatibilities of Zhishi and Baizhu have the function of promoting stomach intestine dynamia and this function is superior to that of using single drug. The best proportion of compatibility is 1: 1 and named Zhizhu Yin. Zhizhu Yin can regulate the dispelling of acetylcholine (Ach) and nitrogen monoxidum (NO) which is a kind of neurotransmitter to adjust the excitation and containment in enteric nervous system (ENS) so as to influence the function of M and D endocrine cell, adjust the content of MTL and VIP which is a kind of gut hormone with function of excitation and containment and regulate gene expression of gut hormone receptor. Thus it can promote gastrointestinal motility and treat FD.
