Objective: To study the inhibitory effects of recombinant adenoviruses carrying the mouse endostatin gene therapy on endometriosis in nude mice. Methods: Replication-defective recombinant adenovirus vector containing mouse endostatin gene was constructed. Its biological activities and expression were surveyed in vitro.Animal model of endometriosis bearing nude mice were established to assay the expression of mouse endostatin and inhibition for endometriosis in vivo. Results: Titer of the recombinant adenovirus was about 5.0×10~9 Pfu/ml.The expression of Endostatin gene was explored in the level of both mRNA and protein. the endometriotic lesions was inhibited in nude mice subject to injection of the recombinant adenovirus when compared to control mice treated with Ad-LacZ. Immunohistochemistry reveals ectopic endometria cell could be stained by Anti-endostatin,indicating that the gene could be expressed in the endometriotic lesions.The microvessel density (MVD) was also decreased significantly ,while apoptotic rate was increased in the treated mice.Conclusion: The recombinant adenovirus can express biologically active endostatin effectively,which inhibits the endometriotic lesions growth by antiangiogenic mechanism.This lays the foundation for the angiogenesis therapy of endometriosis in clinical application.
