PURPOSE Proliferate vitreoretinopathv ( PVR ) is a specific process of the wound healing in the eye, which parallels to general wound repair with three phases of inflammation, cellular proliferation and scarring. The initial factors of the early stage in the wound repair is still unclear . In 1986 Hui established an inflammatory PVR model, which offered us an innovate way to comprehend the mechanism of PVR . The purpose of this study is to observe the 4 proinflammatory cytokines' dynamic changes of TNF-α, IL-1β, EL-8 and IL-6 in macrophage-induced inflammatory PVR model. These 4 cytokines are earlier secreted by macrophages, appear in the initial stage of PVR course and may play mediating roles in the development of PVR. We investigate the levels of the 4 cytokines in supernatants of cultured macrophages, the dynamic changes in the vitreous and retina of the PVR model, and the gene's expressions of the 4 cytokines. These changes may suggest new methods for prevention and treatment of PVR.MATERIAL AND METHODS 1. We collected and purified the homologous activated macrophages from pigmented rabbit's peritoneal cavity after injection of 3% thioglycollate medium. The cell suspension with 5% fetal bovine serum at a concentration of 3.8 × 10~7/ml ~ 5 × 10~7/ml was cultured. The supernatant was extracted at 8h,20h,48h,72h,and 96h respectively. 2.
