PCR methods were used to detect HPV types of the squamous carcinomas of the cervix (SCC), condyloma acuminata (CA) of the cervix and vulva. Reverse-transcription polymerase chain reaction(RT-PCR) methods were used to detect mRNA of c-jun, c-fos, p21 and MDM2. Immunohistochemistry methods were used to detect expression of proteins of c-jun, c-fos, p21, MDM2, HPV16 E6, HPV16 E7 and Ki67. Tunnel methods were used to detect apoptosis of CA of the cervix and vulva and SCC. In vitro, RT-PCR methods were used to detect mRNA of c-jun, c-fos, p21 and MDM2 in transfected keratinocytes by HPV16.The results of PCR showed that HPV6/11 is involved in CA of the cervix and vulva and HPV16 is involved in SCC. The results of RT-PCR showed mRNA of c-jun, c-fos and MDM2 increased more than normal and showed no p21 mRNA. The results of immunohistochemistry showed that expression of proteins of p21 in SCC, CA of the cervix and vulva , was 86.9%, 73.3%, 79.3% respectively; that of c-jun was 55%, 21%, 64.7%; that of c-fos was 63%, 89.5%, 88.2%; that of MDM2 was 78.3%, 73.7%, 58.8%; that of HPV 16 E6 was 50%, 0%, 0%; that of HPV16 E7 was 78.3%, 68.4%, 85.3%; as above, that of Ki67 was distributed in the 1/3~2/3 of the epithelium of CA of the cervix and vulva, and that of Ki67 was in part of the tumour cells of SCC. The results of Tunnel showed that there were very little apoptotic cells in CA of the cervix and vulva, and there were some apoptotic cells in SCC. The results of in vitro test showed that mRNA of c-jun, c-fos, p21 and MDM2 increased in transfected keratinocytes.
