Hepatitis B virus (HBV)infection is a serious disease in China. Approximately 50—70% of the population has been infected and about 10% of them became patients with clinical symptoms. Up to now , mortality of patients with hepatic failure (HF) is quite high. We still have no specific therapy. Stuices recently reported that tumor necrosis factor (TNF) level in HF patients was the highest among those HBV patients at high TNF backgroud. High TNF level is considered to be one of the important factors causing hepatocyte injury or necrosis. Therefore, studies of TNF in HBV infection and searching the TNF inhibitors as theraputic drugs are of practical importance. CHOICE OF TNF MEASUREMENT METHODS4 methods , neutral red, crystal violet , MTT, 3Htymidine release method in which L929 cell was employed as target cell were compared. Results showed that a-mong these (?) methods, crystal violet assay in which target cell densitivity was defined in 3R10? per well and treated with a(?)mycin D should be selected as a simple , rapid, sensitive and stable method. CORRELATION BETWEEN TNF AND HBV INFECTIONCorrelation between Serum TNF Level, Serum IFN, IL -2 and Viral Replication in Hepatitis B Patients.TNF, IFN, IL - L2, ALT and HBeAg in sera of patients were simultaneously determined. Results showed that TNF level of patients with acute hepatitis (AH) and chronic active hepatitis (CAH) were 785. 02±965. 79pg/ml (n = 39) and 1629. 16 ±1183. 50pg/ml(n =11) respectively both higher than healthy controls (165. 53± 98. 53pg/ml ,n= 10). Serum TNF levels of CAH patients were higher than AH patients. Serum TNF levels of patients with positive HBeAg were significantly higher than those with negative HBeAg. Serum levels of IFN and IL—2 were normal in patients with TNF level above 800pg/ml, but low in patients with TNF lower than 20 pg/ml.
