The Study On The Effect And Mechanism Of Electroaccupuncturing "Shenshu" Point And "Huiyang" Point In Rats Model Of Experimental Benign Prostatic Hyperplasia

Objective To probing into the effective method of treating Benign prostatic hyperplasia (BPH) , prove the validity of BPH treated by electroaccupuncturing "Shenshu" point and "Huiyang" point and to study the action mechanism of BPH further which could instruct clinic and optimize the scheme of select points.Methods (1) We made the animal model of BPH with fifty rats, male, by the method of sucutaneously injecting testosterone propionate after castration. (2)Fifty SD male rats were divided into five groups of which each group include ten rats randomly. Of them one group was choused as control group, others were divided into four groups, namely model group done by sucutaneously injecting testosterone propionate, traditional acupuncture group in which rats were acupunctured "Guanyuan" point , "Zhongji" point and "Sanyinjiao" point, medicine (fmasteride) group and electroaccu-puncture group in which rats were electroacupunctured "Shenshu" point and "Huiyang" point. In addition to the control group, the rats in other groups were castrated. After a week, the rats castrated were injected hypodermically testosterone propionate everyday for four weeks, and made the corresponding disposal at the same tine. (3) Observe the changes of weight, exponent and volume of prostates of rats. (4)Observe morphological changes of prostates, measure the epithelial cell height of prostate and the diameter of gland antra under light microscope. (5)Observe the ultrastructural changes of prostate cells of rats under electron microscope. (6) Measure the content of NO in each prostate tissues of rats through the method of "nitric acid deoxidizing enzyme" . (7)Detect the expression of Bcl-2 albumen, Bax albumen and Caspase-3 in each prostate tissues of rats through immunochemical method. (8)Detect the expression Bcl-2 mRNA of each prostate tissues of rats through the method of in situ hybridization (ISH).Results (1)The prostate weight, exponent, volume, the epithelialheight and the diameter of prostate gland antra of rats in model group were higher notably than those in other groups ( P < 0.01 ) . Electroaccupuncturing "Shenshu"and "Huiyang"point which was not significant difference(P>0.05) compared with medicine in respect of minishing volume, can reduce the prostate weight and exponent more obviously than the other two therapy groups (P<0.05 ) . (2)The prostate tissues of Model rats were found obvious hyperplasia in histology and ultrastructure. Electroaccupuncture can relieve the anomaly evidently. Under electronmicroscope the manifestation of early apoptosis was showed in prostate cells of electroaccupuncture and medicine group. ?Compared with model group, Electroaccupuncture and traditional acupuncture had increased markedly the level of NO which decreased more greatly in model rats'prostate than that in contro group (P<0.01 ) . The effect of electroaccupuncture was better than that of traditional acupuncture (P<0.05 ) . @The level of expression of Bcl-2 albumen and Bcl-2 mRNA were higher in model group than that in contro group after treatment in four weeks ( P<0.01 ) , but the level of expression of Bax decreased significantly ( P<0.01 ) , which caused the proportion of Bcl-2/Bax in prostate cells larger markedly than that of rats in control group (P<0.01 ) . In electroaccupuncture group the reasult showed that the expressive level of Bcl-2 albumen and Bcl-2 mRNA became lower, and the expressive level of Bax albumen became higher than that in model group (P<0.01 ) .The effect of electroaccupuncture regulating the proportion of Bcl-2/Bax was better than that of other therapy groups (P<0.05 ) . ?The expression of Caspase-3 in prostate tissue of model rats was lower than that of control group (P<0.01 ) . In electroaccupuncture group the expression of Caspase-3,which was higher than in other groups (P<0.05) was promoted markedly (P< 0.01 ) .Conclusions (pit was effctive for preventing and curing experimental BPH of rats to electroaccupuncture "Shenshu" point and "Huiyang" point,which could decrease the weight, exponent, volume, the epithelial height and the diameter of rats' prostate gland antra, promote the apoptosis, reduce the cell amounts of mesenchymalsmooth muscle and inhibit the hyperplasia of prostate. (2) Electroaccupuncture can counterwork lower urinary tract obstruction (LUTS) of BPH which was led by dynamic factors and static factors, by increasing markedly the level of NO which can loosen up smooth muscle, inhibit the cell multiplication of prostate and induce the apoptosis. (3) Electroaccupuncture can counterwork the damage caused by static factors caused by the hyperplasia of prostate though regulating the expression of Bcl-2 albumen and Bcl-2 mRNA downwards and promoting the expression of Bax albumen and Caspase-3, which could promoted the apoptosis.
