Objective: Investigate the technic of neural stem cells (NSCs) culture. Observe the relationship between the NSCs collection and proliferation. Compare the proliferation and passage capability of NSCs of different source. Observe the effect of cytokine and NSCs injection into injured brain. Methods: The cells isolated from different fetal age and different site were cultured in serum-free medium. The NSCs derived from motor cortex and hippocampus and the cytokine bFGF were injedted into injured motor cortex. Ethology and immunohistochemistry detection were undertake. Results: The NSCs isolated from young fetal age or near the germinal center possess strong proliferation and passage ability. Post brain trauma, intra-cotex injection of bFGF and NSCs could obviously improve motor defect. NSCs orthotopic transplantation could bring about better long-term effect. Conclusion: NSCs possess connatural rule about proliferation and differentiation. The origining of NSCs confine their ability of proliferation and passage. NSCs orthotopic transplantation is a good measure.
