Spinocerebellar Ataxian (SCAs) is a neurodegenerative disorder, characterized by a progregressive atrophy of cerebellum, pons, inferior ol i varis nucleus and loss of neural cells with the prol i feration of neurogl ia. There are a loss of Purkinje cell, a thinning of granular layer, a depletion of dentate nucleus in the cerebellum and a moderately degree loss of inferior olivaris nucleus neurons and a different degree degeneration in the substantia nigra(SN) . external globus pal1idus(GPe) , red nucleus and posterior cord of spine. At present, SCAs are thought that it is a heredity disorder and its pathogenesis is elusive . It has not effective approach on the clinic treatment. With the development of neural cells transplantation .several investigators brought forward the strategy on the treatment of SCAs. In resently years, the evidence of study shows that there is neural stem cells (NSCs) characterized by a multipotentiality of differentiation and a capacity for self-renewal in the developing mammauna nervous system.On certainly condition, it can be proliferated and be induced differentiation that can give rise to different neural cells in the central neural system (CNS) in specified way . Potential uses of NSCs in repair include transplantation to repair missing cells in the treatment of the trauma of CNS and the neurodegenerative disorder(include SCAs) and the activation of endogenous cells to provide "sel f-repair". It is becoming one of the latest advances in the treatment of SCAs.For the study of experimentation, we have to establish steady animal model of SCAs. First, Rats receive a consequent intraperitoneal injection of 3 acetylpyridine. harnaline and nicotinamide at intervals. After leion surgery, animals were tested the gait width beheavior every week and were checked the morphology change of cerebellum, mesencephalon and pons. Just at the day of surgery , rats exhibited disorder of balance and abnormal gait. During (he following week,the symptom was progresive aggravating. Soon after, the symptom was reliefing. Since tow weeks, disorder of balance and abnormal gait has been steadily. The light loss of cerebellum Pukenje cell, neurofibra confused of the pons and mesencephalon and the obervious depletion of inferior olivaris nucleus are major morphology changes. So this animal model is fit for the study of transplantation.In order to obtain the enough neural precursor with the potential capacity of differentiation, Fetal cerebellum was microdissected from fetal human (E12 weeks) under sterile conditions .gently triturated into a cell suspension and then plated flasks in expansion media(F12/DMFM.
