| Objective: To explore the various GGTmRNA types of HCC cancerous tissues, noncancerous tissues from livers with HCC, liver tissues without HCC. To investigate the relationship of the change of GGTmRNA types and HCC. To explore the value of analysis of GGTmRNA types in monitoring hepatocytes carcinogenesis and early diagnosing of HCC. To study the clinical value of detecting GGTmRNA in peripheral blood in searching for cancer cells in HCC patients blood. To explore the new method for monitoring dissemination of HCC so as to provide theoretical foundation for the selection of the treatment manners of HCC.Methods: The three specific primer sets for RT-PCR corresponding to the 5'-noncoding human GGTmRNA were prepared. The GGTmRNA types in cancerous tissues and noncancerous tissues from livers of 65 patients with HCC, types in the noncancerous tissues from 13 patients with other tumor metastatic to liver, types in tissues from 108 patients with benign liver diseases, types in tissues from 6 normal control persons were checked using RT-PCR. Various GGTmRNA types in tissues of various liver diseases were investigated. In order to explore the value of analysis of GGTmRNA types in monitoring hepatocytes carcinogenesis, we toke a 12-monthfollow-up study for 86 patients with chronic hepatitis or liver cirrhosis or alcoholic liver disease, in whose liver tissues GGTmRNA-H were found. Particular GGTmRNA type in HCC was selected serving as the specific target RNA of HCC cell in peripheral blood. The relationship of specific HCC GGTmRNA type in peripheral blood and HCC tumor size, intra- and extra-hepatic metastasis, portal vein thrombosis was analyzed, so that the feasibility and specialty of the method using specific GGTmRNA type to search for HCC cell in peripheral blood were investigated. In order to investigate the value of analysis of GGTmRNA-H and AFP of peripheral blood in predicting metastasis of HCC, we toke a 6-month follow-up study for 41 HCC patients without portal vein and/or extrahepatic metastasis.Results: The main GGTmRNA type in HCC tissue was GGTmRNA-H type, with the positive rate of 93.8% , while the positive rate of GGTmRNA-F type was only 49.2%. The main types in noncancerous tissues of HCC were GGTmRNA-H (81.5%) and GGTmRNA-F types (96.9%). The type in the tissues of benign liver diseases was mostly GGTmRNA-F type (100%), while the GGTmRNA-H type was seldom found. GGTmRNA type in noncancerous tissues of other tumor metastatic to liver is similar to the that in benign liver diseases and normal control persons, GGTmRNA-P type was seldom detected. Through statistic analysis, the positive rate of GGTmRNA-H type in HCC cancerous tissues was strikingly above the one in tissue of benign liver diseases and normal control persons (P<0.01) while the positive rate of GGTmRNA-F in HCC cancerous tissues was below the one in tissues of benign liver diseases and normal control persons (P<0.01). The positive rate of GGTmRNA-H type in noncancerous tissues of HCC was significantly higher than that in benign liver diseases and normal control persons (P<0.01), while the positive rate of GGTmRNA-F in these tissues were similar (P>0.05).
