Objective: To investigate the relationship between the expression of Survivin protein, Survivin mRNA, p27 protein, p27 mRNA, PTEN protein and KAI-1/CD82 protein and tumor cell invasion, metastasis in hepatocellular carcinoma(HCC). Methods: Tissue microarrays were constructed, which contained HCCs, para-carcinoma tissues, distant-carcinoma tissues, intrahepatic metastasis tissues and normal liver tissues. The expression of Survivin protein, p27 protein, PTEN protein and KAI-1/CD82 protein were evaluated by immunohistochemical PV-9000 method in 141 HCCs, 128 para-carcinoma tissues, 97 distant-carcinoma tissues and 17 normal liver tissues. The expression of Survivin mRNA and p27 mRNA were evaluated by in stiu hybridization in the same groups. Further more, the expression of KAI-1/CD82 protien was analyzed in 36 cases of HCC primary lesions and corresponding metastasis lesions.Results:1,Tissue microarrays constructed manually had a good quality. The mean fall rate was 5.27% and the mean observation rate was 97.40%.2,Comparing with the para-carcinoma tissues,distant-carcinoma tissues and normal liver tissues, the expression of Survivin protein, Survivin mRNA and p27 protein in HCCs were significantly increased, while the expression of PTEN protein and KAI-1/CD82 protein in HCCs were significantly reduced. The expressions of all markers showed no difference in para-carcinoma tissues, distant-carcinoma tissues and normal liver tissues.
