To Establish Animal Model of Heart-Qi Deficiency Syndrome by The Method of Comparative Medicine Objective: To establish animal model of heart-qi deficiency syndrome in SD,Wistar,WKY and KM,BALB/c by two methods of swimming or running. By comparative study, we can screen out appropriate method and strain of replicating model, and put forward diagnosis standard of animal model of heart-qi deficiency syndrome. Methods: Every animal in experiment group was forced to load-carrying swim (or run) with slimming until exhausted every day. As deficiency syndrome appeared at the end of experiment, animals were ig propanolol of 24mg/Kg and lasted for 4 days. Measuring indexes as model completed, and by ig supplement qi drug to validate the model. Results: These two methods of replicating model can make both rats and mice deficiency and restrain their heart function. And decreasing cAMP and SOD of rats and SOD of KM. Running method increases MDA of SD and KM and ANP of SD significantly. The results of cytokines show that both of these two methods of replicating model can depress immune function of these two kinds of mice. There is no evident abnormity in pathologic sliceof rats and mice. The results of electron microscope indicate thatthere is different damage in ultrastructure of cell of cardiac muscle. The results of gene chip suggest that damage of vascular endothelium is a main mechanism resulting in heart -qi deficiency syndrome. Conclusion: Both of these two methods of replicating model can reproduce heart -qi deficiency syndrome model on many strains, which is according with theory of TCM, and swimming method is apt to be spread. As for model animal, mice is not feasible, and SD, Wistar are suitable. WKY is not apt to be spread for expensive price. The diagnosis standard of animal model of heart-qi deficiency syndrome is: deficiency appearance, depressed heart function, decreased cAMP and SOD, increased MDA and restrained immune function. ANP could increase in serious case.
