Objectives Osteoporosis is one of the most common old-age diseases attracting people's focus. More and more evidence showed that heredity factors is closely related to the change of bone mineral density (BMD), which is controlled by multiple mini-effect gene. This study is to explore the relationship between the multi-gene type of estrogen receptor (ER), vitamin D receptor (VDR) and BMD, and the multi- gene type of ER and VDR effect on osteoporosis in healthy menopause women. Methods A total of 204 healthy menopause women without blood relationship were enrolled. Dual energy X-ray absorptiometry (DEXA) was used for detecting BMD in lumbar and thighbone, and polymerase chainreaction-restriction fragment length polymorphism (PCR-RPLP) for detecting gene type of ER and VDR With X2 test, we analysis the relationship between the polymorphism of ER, VDR and HMD in menopause women. Results Of the 204 healthy menopause women, according to the BMD detection results, 71 were normal, 74 of BMD decrease, and 59 osteoporosis; according to ER gene type, 40 cases were PP type, 98 Pp type, and 66 pp type; and according to VDR gene type, 188 cases were bb type, 16 Bb type, and no detection of BB type. For ER gene type, the occurrence of osteoporosis was highest in women with PP gene type, and lowest in women with pp type. For VDR gene type, the occurrence of osteoporosis is higher in women with bb type (31%), which has a significant difference compared with that in women with Bb type (6%, P<0.01). Combined analysis of VDR-ER multi-type showed that BMD was remarkably lower in cases with bb-PP typethan that in Bb-pp type (P<0.01). Conclusion bb-PP gene type cases have a higher risk rate of osteoporosis. In molecular biology, bone decrease is realistically related to the prevention of the occurrence of osteoporosis. To exclude high-risk population using molecular heredity and to detecting some easy infected of osteoporosis can provide theory for further genatic interference of osteoporosis.
