Effects and Mechanism of Active Site from Reinforcing Kidney Chinese Herbs on Bone Formation Ph.D candidate: Yaqiong Chen Supervisor: Prof. Shuguang Wu The Research Institute of Pharmacology, The First Military Medical University Abstract Reinforcing kidney Chinese herbs is functioned similarly as estrogen when preventing and treating osteoporosis, and partly as antiestrogen in reproductive system. We have been devoting to research on whether estrogen receptor has brought about these effects. This study, following the effects of estrogen, focused on active site from Reinforcing kidney Chinese herbs (AS-RKCB), which was evaluated from such four aspects as whole body, cells, molecules, and genes. By testing bone mineral density (BMD) and behavior of bone biomechanics in ovariectomized (OVX) mice, and biochemical markers of bone formation in OVX mice and osteoblasts, and by in situ hybridization technique, the effects and mechanism of AS-RKCB on enhancing in vivo and in vitro were proved, mechanism of the receptor promoting effect of estrogen was indicated, and the expression of core binding factor a I (cbfal) was illustrated by regulating osteoblastic cells, which had effect on ?? secretion of osteocalcin(OC) in the lower gene. Results: First, AS-RKCB promoted the BMD in mouse抯 thigh, strengthened the nature of the structural mechanics, reduced bone turnover rate, and effectively prevented osteoporosis induced by OVX. Second, less estrogen-like effect of AS-RKCB in reproductive system could promote uterine wet weight and keratinization rate by mice~ s vagina cell smear, but the role is 10 per cent lower than that of estradiol. Third, AS-RKCB, whose effect mechanism was activated through ER, promoted osteoblastic proliferation and activation of alkaline phosphatase by time-depended and doze-depended way; and mechanism of AS-RKCB promoting osteoblast to secrete of OC was irrelevant to receptor activation. Finally, AS-RKCB promoted the expression of core binding factor a 1 (cbfal) by regulating function of osteoblast, which enhanced the secretion of OC the product of lower gene.
