Objectives:This study was to investigate the conduct effects of plarycodon root and/or bidentate achyranthes root of Xuefu Zhuyu decoction via peoniflorin pharmacokinetics and organ contribution in normal and blood stasis rodents.Methods:KM mice (n=48, half male and half female) were randomly divided into normal group (n=24) and blood stasis group (n=24). After 12 h fasting with free access of water, the blood stasis group had been injected with 25% dextran in T500 saline (20 mL/kg) via tail vein. After 10 min of injection, the mice were intubated with Xuefu Zhuyu decoction (13.742 mg/kg peoniflorin; 6 mL/kg) in the presence or absence of plarycodon root and/or bidentate achyranthes root (n=6 in each decoction). The blood, brain, heart, liver, spleen, lung, and kidney samples were collected and determined the peoniflorin concentration via HPLC.SD rats (n=48, half male and half female) were randomly divided into normal group (n=24) and blood stasis group (n=24). After 12 h fasting with free access of water, the blood stasis group had been injected with 5% dextran in T500 saline (2 mL/kg) via tail vein. After 30 min of injection, the rats were intubated with Xuefu Zhuyu decoction (11.452 mg/kg peoniflorin; 5 mL/kg) in the presence or absence of plarycodon root and/or bidentate achyranthes root (n=6 in each decoction). The orbital venous plexus blood was sampled at 5,10,15,20,30,45,60,120, and 240 min after administration in normal group and 10,15,20,30,45,90,150, and 240 min after administration in blood stasis group. The blood peoniflorin concentration was investigated by HPLC.Results:Compared with Xuefu Zhuyu decoction in the absence of plarycodon root and bidentate achyranthes root, (1) Xuefu Zhuyu decoction in the absence of plarycodon root contributed the peoniflorin distribution in the mice lung, kidney, heart, liver, and spleen, and enhanced ti/2z of peoniflorin in rats in blood stasis group. (2) Xuefu Zhuyu decoction in the absence of bidentate achyranthes root increased the peoniflorin level of kidney in normal mice. It also increased the peoniflorin levels of lung and kidney in mice, but not affected the pharmacokinetics of peoniflorin in rats of blood stasis group. (3) Xuefu Zhuyu decoction (presence of plarycodon root and bidentate achyranthes root) enhanced the peoniflorin concentration of kidney, contributed the peoniflorin distribution in the heart, liver, spleen, and lung in mice, and increased Ke of peoniflorin in rats in blood stasis group. Moreover, it increased the ln(Cmax) and Ke, and decreased t1/2 of peoniflorin in normal rats.Conclusions:(1) This study provided the evidence that the conduct herbs of Xuefu Zhuyu decoction, plarycodon root and bidentate achyranthes root, guided peoniflorin to the organs, increased peoniflorin distribution in the related organs, and elevated the treatment functions, which is the mechanism of conductor. (2) Plarycodon root might diffuse the lung qi and regulate the dynamic. Qi movement improved the blood circulation. Therefore, plarycodon root contributed the peoniflorin distribution in lung and kidney. (3) Bidentate achyranthes root might increase the peoniflorin distribution in lung, kidney, heart, liver, and spleen under blood stasis constitution, which may related to its function, includes to improve the blood and meridiam circulation and to transform the stasis...
