Helicobacter pylori (H. pylori) infection is the main cause of chronic gastritis and digestive ulcer, especially it can lead to gastric carcinoma. The world health organization has reported that Helicobacter pylori belongs to the first class factor leading to carcinoma. The cure rate of chronic gastritis and digestive ulcer, especially gastric carcinoma is very low. At present, the hydrochloric acid inhibitor and two or three kinds of antibiotic are used in the method of eliminating H. pylori. However, the Helicobacter pylori infection is general, there are more and more Helicobacter pylori bacterium resistance to the action of medicine and the medicine is very expensive. Therefore, how to prevent and cure H. pylori infection is a very important task of clinical treatment. In recent years, people progressively recognize that direct mucous membrane immunity can protect persons against H. pylori infection. Vaccine is the most effective method to prevent and cure Helicobacter pylori infection. By using transgenic plants, plant storage organs could serve as factories to produce protein of biotechnological interest. Oral plant vaccine makes people in the world prevent and cure Helicobacter pylori infection more convenient and more economic. Oral plant vaccine has several advantages, such as cheap, easy storage, no syringe needle and injector required etc.In this study, we successfully constructed the plant expression cassette with HspA and transformed it into tobacco. 14 regernerated tobacco plants were obtained in selective medium. 11 independent transformants were positive by PCR analysis, showed the presence of the HspA gene in all PCR positive transformants, indicating that the foreign HspA gene was integrated into tobacco genome. Leaves were taken from independent transformants for RNA extraction. We performed RNA isolation and northern blot hybridization.
