Purpose:In this study,a mouse skin photoaging model was established by UV irradiation,and the effect of Taohong Siwu Decoction on antioxidant Nrf2/Ho-1 signaling pathway during UV induced photoaging in mouse skin was investigated.Material and method:Randomly divide 60 mice into 4 groups(n=15).Except for the blank group,all other groups were exposed to ultraviolet(UVA+UVB)light for 7 weeks,2hours per day.After the end of illumination,medication intervention was given for 4 weeks.The blank group was given 0.4 ml of 0.9% physiological saline per day,the model group was given 0.4 ml of 0.9% physiological saline per day,the traditional Chinese medicine group was given 0.4 ml of 1.23g/ml Taohong Siwu Tang per day,and the western medicine group was given 0.4 ml of vitamin E per day.After the experiment,samples were collected,including mouse eyeball blood and mouse back skin tissue.HE staining was used to observe the morphology of the back skin tissue of each group of mice,Western blot was used to detect the expression levels of Nrf2 and Bach1 proteins,immunohistochemistry was used to detect the relative quantification of Nrf2 and Bach1 proteins,and enzyme linked immunosorbent assay(ELISA)was used to detect the concentration of HO-1 in serum.Results:1.Preparation of photoaging model: The irradiation dose meets the UV dose requirements of skin photoaging animal experiments.The cumulative UVA light source has reached261.79J/cm2,and the cumulative UVB light source has reached 6.54J/cm2.After 7 weeks of UV irradiation,mice were observed to exhibit loss of appetite and mental exhaustion.The naked eye could see that the skin wrinkles on the back of the mice deepened,the skin was rough and dry,the skin atrophied,and the skin tone deepened;HE staining showed that the skin tissue of the model group mice became significantly thicker than that of the blank group,with a large number of inflammatory cells infiltrating the dermis,and collagen fibers arranged dispersedly and broken,indicating the successful establishment of a UV induced skin photoaging model in mice.2.Enzyme linked immunosorbent assay(ELISA)was used to detect the concentration of HO-1in serum.The results showed that compared with the blank group,the concentration of HO-1in the model group decreased(P<0.05);while compared with the model group,the concentration of HO-1 in the traditional Chinese medicine group increased(P<0.05);Compared with the model group,the concentration of HO-1 in the western medicine group increased(P<0.05).3.Western blot analysis of Nrf2 and Bach1 protein showed that compared with the blank group,the expression of Nrf2 protein decreased(P<0.05)and the expression of Bach1 protein increased(P<0.05)in the model group‘ Compared with the model group,the expression of Nrf2 protein in the traditional Chinese medicine group and the western medicine group increased(P<0.05),while the expression of Bach1 protein decreased(P<0.05).4.Immunohistochemical detection of Nrf2 and Bach1 protein showed that compared with the blank group,the expression of Nrf2 protein decreased(P<0.05),while the expression of Bach1 protein increased(P<0.05)in the model group;Compared with the model group,the expression of Nrf2 protein in the traditional Chinese medicine group and the western medicine group increased(P<0.05),while the expression of Bach1 protein decreased(P<0.05).Conclusion:1.Taohong Siwu Decoction can significantly improve the aging caused by photoaging of mouse skin.2.Taohong Siwu Decoction can activate the antioxidant Nrf2/Ho-1 signaling pathway in the body,promote the expression of Ho-1 by inhibiting the expression of Bach1,improve the content of antioxidant enzymes in the body,eliminate oxygen free radicals,and protect the body from oxidative stress induced light damage.
