Objective: To observe the repair effect of Taohong Siwu Decoction on cartilage injury model,observe and explore the possible mechanism of BMP-3 signal pathway in repairing cartilage injury using modern molecular biological technology,so as to provide evidence for t he clinical use of Taohong Siwu Decoction.Methods: Forty healthy New Zealand rabbits,weighing2.5~3kg/rabbit,were randomly divided into two groups after adaptive feeding for one week,the blank group(10 rabbits)and the model group(30 rabbits).In the modeling group,different degrees of cartilage injury models were made in the left knee joint of the same rabbit,the femoral knee joint groove and the medial,lateral and median of the femoral joint surface.Antibiotic anti-infection treatment was given for 3 days after operation,and the wound condition was observed.There was no obvious sign of infection.On the 4th day after operation,they were divided into traditional Chinese medicine group(Taohong Siwu Decoction group),western medi cine group(glucosamine group)and model group(saline group)according to the method of random number table,and the corresponding groups of Taohong Siwu Decoction,glucosamine,and saline were administered by gavage respectively.At 4 and 6 weeks after g avage,5 rabbits were randomly selected from each group.After taking samples,the model was subjected to gross observation,ELISA,Western Blot and q-PCR detection and statistical analysis.Results: The traditional Chinese medicine group,the western medicine group and the model group can well tolerate the experimental operation process,and there is no obvious infection after the operation.They can move,eat and drink normally,and there is no death.The results of enzyme-linked immunosorbent assay,BMP-3 protein expression and q-RCR showed that the expression level of traditional Chinese medicine group,western medicine group and model group were higher than that of the blank group;The difference between the Chinese medicine group and the western medici ne group was statistically significant(p<0.001).The WB results showed that the expression of BMP-3 protein in the Chinese medicine group was lower than that in the western medicine group at the fourth week,and the difference was statistically significan t(p<0.001).Conclusion: Taohong Siwu Decoction can promote the repair of cartilage injury by down-regulating the expression of BMP-3 signal in rabbit cartilage injury model.
