Objective:Document the prescription and medicinal flavor,clear prescription source,dosage composition,origin and origin of 15 batches of medicinal materials and decoction parts,mainly including thin layer identification and content determination and establish fingerprint map,then study the preparation material benchmark,establish the material benchmark,determine the content of index composition,and finally explain the transfer relationship of "medicinal material-decoction piece-material benchmark".Method:1.According to the principle of "original prescription,original dose and original preparation method ",the 1.consulted ancient literature and modern clinical medical records,combined with modern clinical application and 2020 edition of Chinese Pharmacopoeia,determined the prescription source,prescription composition,prescription dose,prescription efficacy and application,and determined the origin,medicinal location and processing method of prescription medicinal materials.2.According to the results of the literature study,6 medicinal materials were selected from 3 producing areas and 15 batches,and according to the2020 edition of Chinese Pharmacopoeia,the quality of medicinal materials and pieces were checked,including thin layer identification method,content determination method,and the HPLC fingerprint of each medicinal material and pieces were established.3.The 15 batches of 6 kinds of traditional Chinese medicine in the prescription were completely randomly divided into groups,and the decoction was made strictly according to the prescription method.By using high performance liquid chromatography(HPLC)to establish the fingerprint of the material benchmark,to calculate its similarity,to determine the number of common peaks,to detect the content of the index components in the material benchmark,and to calculate the transfer rate of the index components of the medicinal materials-pieces-material benchmark.4.Methodological review of the above methods,including linear relationship,precision,stability and reproducibility.Results:1.Peach four soup from Zong Jin Jian,prescription of ripe ground,angelica,white peony,xiong,peach,safflower,rear Chinese medicine flavor changed from cooked to raw,including the processing process of decoction pieces: raw,angelica,red for wine wash,white peony for wine frying,peach tip research mud,Chuanxiong net raw production.Among them,11.19 g,Angelica 14.92 g,Bai Peony 5.60 g,Chuanxiong 3.73 g,Peach 3.78 g,red flower 3.73 g.Prescription frying method is adding pure water 520 m,soak30min,martial fire heating 15 min,classical thermal heating 20 min,average frying time is 35 min,3 layers of gauze filter filter liquid 410 ml,freeze drying.Ingredients: Weiqing Prefecture,Wuqing County,Henan Province;Angelica is Minxian County,Weiyuan County and Longxi County,Gansu Province;Chuanxiong is Dujiangyan,Sichuan Province,Bai Peony is Bozhou,Anhui Province and Hangzhou,Zhejiang Province;Taoyuan is Chenzhou Prefecture and eshan County in the southeast of Henan Province;and safflower is Xinjiang region.2.The index component contents of the six traditional Chinese medicines and decoction pieces were as follows:(1)rehmannia glycoside D(rehmannia glutinosa): 0.239± 0.024%;(2)ferulic acid(Angelica sinensis): medicinal materials 0.169± 0.024%;(3)Paeoniflorin(Paeonia lactiflora): medicinal materials 2.184± 0.1112%;(4)ferulic acid(Ligusticum chuanxiong):medicinal materials 0.12± 0.0065%;(5)amygdalin(peach kernel): medicinal materials 2.335± 0.1375%;Decoction pieces 3.0979± 0.0501%(6)Hydroxy Safflower Yellow A(Safflower): medicinal materials 1.682±0.123%;Decoction pieces 2.0527± 0.231%.There was no statistical significance in comparison of index component content between producing place and batch(P BBB 0.05).It indicated that the content of index components between 15 batches of medicinal materials and decoction pieces from 3 producing areas was relatively stable.3.The material reference of Taohong Siwu Decoction was prepared.The content of index components was as follows:(1)rehmanniin D: 0.15±0.026%;(2)Ferulic acid: 0.497± 0.31%;(3)paeoniflorin: 0.673± 0.257%;(4)amygdalin 1.247± 0.218%;(5)Hydroxy safflower yellow pigment A: 0.341±0.24%.Compared with the content in the decoction pieces,the transfer rates were:(1)rehmannia glycoside D: 34.58± 0.2258%;(2)Ferulic acid: 20.315±1.806%;(3)paeoniflorin: 29.856± 3.134%;(4)amygdalin: 40.26± 0.025%;(5)Hydroxy safflower yellow pigment A: 16.85± 0.658%.The extraction rate of Taohong Siwu decoction was 36.32± 1.94%.4.The HPLC fingerprint of Taohong Siwu Decoction was established,and the similarity between 15 groups reached more than 90%,which proved that the similarity between each producing area and each batch was high,and the preparation method of Taohong Siwu Decoction material reference was stable and repeatable.Seventeen common peaks were identified.The third peak was rehmanniin D,the eighth peak was hydroxyl safflower yellow A,the eleventh peak was amygdalin,the twelfth peak was paeoniflorin,and the thirteenth peak was ferulic acid.5.The above methods have good linear relationship,instrument precision,method repeatability and sample stability,and the relative peak area and relative retention time RSD are less than 3%.Conclusion:1.there was no significant difference between 15 batches of 6 kinds of medicinal materials in 3 producing areas of Taohong Siwu decoction,which could be used as the standard medicinal material standard of Taohong Siwu decoction.2.the stability and reproducibility of Taohong Siwu decoction,it can be used as the preparation method of its material benchmark.
