Purpose : This study is based on the modern literature in the network database,systematically mining the medication rules of post-stroke insomnia,and based on the statistical results,using the research methods of network pharmacology to predict the potential material basis and mechanism of high-frequency medication for post-stroke insomnia,in order to pave the way for subsequent experimental research.Material and method:Retrieve relevant documents in the databases of CNKI,Wanfang and Vip,and based on the SPSS statistical software,count the frequency of the medication of insomnia after stroke and the frequency of the four qi,five flavors and meridian tropism,and explore the core drug combination and possible new formula combination.The action mechanism of high frequency medication for insomnia after stroke was analyzed by using the method of network pharmacology.The drugs and disease targets were collected through BATMA database,Dis Ge NET database,Gene Cards database,OMIM database and other databases,and then the intersection of disease and drug targets was obtained by using the Wayne map.On this basis,PPI network analysis,GO enrichment analysis and KEGG enrichment analysis are carried out,and the results are visualized.Results:1.This study included 101 prescriptions of traditional Chinese medicine.It was found that the commonly used drugs for the treatment of insomnia after stroke were poria cocos,semen ziziphi spinosae,liquorice,etc.The four qi,five flavors,meridian tropism,moderate cold,warm,sweet and bitter,liver tropism and heart meridian tropism of the medicine are mostly used.The drug pairs with higher frequency are "Poria cocos,licorice","licorice,jujube seed",etc.Through cluster analysis,four potential new combinations are mined.2.Based on network pharmacology,it has been predicted that the key active ingredients of "Poria cocos sour jujube kernel" are ergot amine,betulinic acid,maltose acid,etc.These active ingredients mainly act on targets such as CACNA1 C,CACNA1D,IL1 B,IL6,TNF,etc.through neural active ligand receptor interaction,cyclic adenosine monophosphate signaling pathway,calcium signaling pathway,serum hormone synaptic pathway,etc.to treat insomnia after stroke.Conclusion:1.After stroke,there are more deficiency and blood stasis.The drugs are mainly used to nourish the heart,calm the nerves and promote blood circulation.Because the stroke patients have been high for many years and lack of kidney essence,they often choose sweet,cold and mild products to nourish yin and reduce fire.The drugs are mostly directed to the liver and heart meridians,which can help the blood stasis disperse and promote the recovery of limb function;In addition,the liver stores blood and the heart stores spirit.The liver blood should be able to replenish the heart blood and make the mind stable to treat insomnia.2.The mechanism of the intervention of "Poria cocos sour jujube kernel" on post stroke insomnia is closely related to metabolic pathways such as the interaction of neural active ligands receptors,cyclic adenosine monophosphate signaling pathway,calcium signaling pathway,and serotonin synaptic pathway.
