Purpose:Apply network pharmacology and data mining methods to explore the medication rules of traditional Chinese medicine for diabetic retinopathy,extract high-frequency drug pairs,predict the effective components,target proteins and related pathways for the treatment of diabetic retinopathy,and construct chemical components-target proteins-the signal pathway map and the protein-protein interaction network map provide more clinical evidence for the treatment of diabetes mellitus with traditional Chinese medicine.Material and method:1.Search CNKI,VIP,Wanfang and other electronic journal databases,screen documents according to the inclusion and exclusion criteria,the time is limited from 2000.01.01 to2020.12.31,and extract the publication time,author,Chinese medicine prescription and other information of relevant documents Establish a database and import the TCM inheritance auxiliary platform to analyze the frequency of related drugs,nature and flavor,meridian tropism,and combination of new prescriptions.2.The TCMSP database was used to retrieve information on the active ingredients and related protein targets of high-frequency drug pairs "Astragalus-Salvia miltiorrhiza",and the potential targets of diabetic retinopathy were retrieved in the OMIM、Gene Cards database.The drug targets and disease targets were intersected and imported into the STRING database to construct a protein-protein interaction(PPI)network.The key targets were screened by topological analysis using Cytoscape software.The chemical composition-target protein-signaling pathway network map was constructed by Cytoscape software.GO and KEGG enrichment analysis was performed on the intersection target proteins using the Metascape database.Results:1.A total of 204 prescriptions were finally included in the literature,involving 193 drugs,with a cumulative frequency of 2285 times.There were 15 drugs with a frequency of more than 40 times,including Astragalus membranaceus,Salvia miltiorrhiza,Rehmannia glutinosa,Panax notoginseng and medlar,with a frequency of more than 80 and a frequency of more than 40%.Drugs for tonifying deficiency ranked first,followed by heat clearing drugs,drugs for promoting blood circulation and removing blood stasis,hemostatic drugs,surface relieving drugs and drugs for promoting water and dampness,which were used more than 100 times.The four natures of drugs are mainly cold,and sweet drugs account for the largest proportion among the five flavors.Most drugs belong to the liver and kidney channels.The frequencies of Salvia miltiorrhiza and astragalus,Rehmannia glutinosa and astragalus,Rehmannia glutinosa and Salvia miltiorrhiza were more than 60 times.26 core drug combinations and 13 potential new prescriptions were obtained by entropy cluster analysis.2.A total of 85 effective chemical constituents were obtained from "Astragalus-Salvia miltiorrhiza",including 20 Astragalus and 65 Salvia miltiorrhiza.There are 256 corresponding target proteins.4249 disease protein targets were obtained.There are 133 target genes of "Astragalus-Salvia miltiorrhiza" and diabetic retinopathy.The top three core components of "Astragalus-Salvia miltiorrhiza" in the treatment of Dr include quercetin,luteolin and kaempferol.The 29 main targets include AKT1,TNF,TP53,IL6,CASP3,EGFR,IL1 B,JUN,STAT3,MMP9,Myc,HIF1 A,PTGS2,ESR1,PPARG,HSP90AA1,CCND1,CXCL8,PTEN,FOS,ERBB2,MMP2,CCL2,IL10,NOS3,EDN1,serpine1,PPARa and APP.The pathway is rich The results include AGE-RAGE signal pathway,tumor necrosis factor signal pathway,HIF-1 signal pathway,VEGF signal pathway and NF-κB signaling pathway,apoptosis signaling pathway,insulin resistance signaling pathway,etc.Conclusion:1.The pathogenesis of diabetic retinopathy is mainly due to yin deficiency and dryness heat and blood stasis blocking the collaterals.The treatment is mainly from the liver and kidney,supplementing qi and nourishing Yin,clearing away heat and improving eyesight,activating blood circulation to dredge collaterals,and promoting dampness and dampness.Clinical application should be strictly based on syndrome differentiation and select prescription and medication.2.In this study,the network pharmacology research method was used to conduct preliminary research on the effective components,target proteins and pathways of "Astragalus-Salvia miltiorrhiza" in the treatment of DR.The main components of "Astragalus-Salvia miltiorrhiza" in the treatment of DR may be quercetin,luteolin,kaempferol,etc.,and may act through potential targets such as AKT1,TNF,IL6,CASP3,IL1 B,etc.,and then affect the AGR receptor of diabetic complications.Signaling pathway,tumor necrosis factor signaling pathway,HIF-1 signaling pathway,VEGF signaling pathway,NF-κB signaling pathway,apoptosis and other signaling pathways play a regulatory role in response to trauma,vascular development,apoptosis signaling pathway,Response to growth factors,negative regulation of cell population proliferation and other biological processes,play a therapeutic role.
