Objective: Through data mining,Professor Luo Zhijuan’s prescription rules and treatment experience in treating infertility in elderly women were studied;Network pharmacology was used to analyze the possible pharmacological action components,pharmacodynamic targets,signaling pathways,etc.of the core formula,and explore their mechanism of action.Methods: 1.Using the TCM inheritance assistance platform system V3.0 to mine the medical cases of Professor Luo Zhijuan from September2020 to January 2023 to identify and treat elderly infertility patients,the content includes: infertility type,drug frequency,sexual taste,attribution,efficacy,association rules,cluster analysis,analyze the medication characteristics of Professor Luo Zhijuan in the treatment of elderly female infertility,and refine the core formula.2.Relying on the data mining results,the mechanism of action of Professor Luo Zhijuan’s core formula for the treatment of infertility in elderly women was explored by using network pharmacology technology.The effective chemical components and targets of the core group were screened through TCMSP and BATMAN databases;Obtain disease targets with "Infertility" as the keyword through the Gene Cards,OMIM,and Dis Ge NET databases;R4.2.1 was used to analyze the core group targets and infertility targets to obtain the potential targets of core formula for the treatment of infertility,and the network diagram of "traditional Chinese medicine-active ingredient-intersection target" was constructed by Cytoscape 3.7.2 to screen the core components.The STRING platform was used to draw PPI networks,and Cytoscape3.7.2 software was used for topological analysis to mine core targets;GO enrichment analysis and KEGG enrichment analysis were performed on intersection target genes by DAVID;and "pathway-target gene" network diagram was constructed by Cytoscape 3.7.2.Molecular docking technology was used to verify network pharmacology results.Results: 1.A total of 1070 prescriptions were included in this study,involving 196 flavors of traditional Chinese medicine,with a total frequency of 14543 times,and the top 10 drugs were licorice,dodder seed,angelica,codonopsis,cooked ground yellow,deer antler cream,yam,goji berry,eucommia,and Chuanxiong;The four qi are mainly flat and warm;The five flavors are mainly sweet;Most of the menstruations belong to the liver meridian,kidney meridian,and spleen meridian;The efficacy is mainly to replenish deficiency,followed by blood circulation and stasis;Based on the support degree of 408,the correlation analysis with a confidence level of 0.8prompts: "cooked ground yellow-dodder seed" and "deer antler cream-dodder seed" are high-frequency drug pairs,and "dodder seed,deer antler cream,goji berry,cooked ground yellow,and codonopsis" are the core groups.Five types of prescriptions were obtained through cluster analysis,(1)peach kernel-safflower-angelica-chuanxiong-citrus husk-lutong;(2)Dodder seed-Codonopsis-Duzhong-Poria-Baishu-angelica;(3)Codonopsis-dodder-raspberry-goji berry-angelica-privet;(4)Dodder seeds-deer antler cream-yam-cooked ground yellow-goji berries-dogwood;(5)Peony peel-poria-yam-ze epsom-dogwood-cooked ground yellow.2.58 effective chemical components of the core formula were obtained through network pharmacology technology,and 216 corresponding targets were obtained;4643 disease targets were obtained;150 potential targets were initially obtained by intersection ratio;Through the construction of protein interaction network and topological analysis,the core active chemical components and core targets were obtained.GO enrichment analysis yielded374 biological processes,which were mainly related to RNA polymerase II promoter transcription,gene expression,and positive regulation of transcriptional DNA template.There were 36 cell components,mainly related to macromolecular complexes,extracellular space,chromatin,cytoplasmic sol,nucleoplasm,etc.There are 79 molecular biological functions,mainly related to binding enzymes and binding of the same protein.KEGG enrichment analysis yielded 287 pathways,which were mainly related to cancer cell pathways,AGE-RAGE signaling pathways,and interleukin-17 pathways in diabetic complications.The molecular docking results showed that the binding energy between key molecules and core target proteins was less than-5.0kca/mol,and the docking efficiency was good.Conclusions: 1.Professor Luo’s core prescriptions for the treatment of infertility in elderly women are dodder seed,codonopsis,deer antler cream,goji berry,cooked rehmania,with "tonifying the kidney" as the main treatment principle,with liver thinning,spleen strengthening,blood circulation and stasis.2.The core group may achieve the therapeutic purpose of antioxidant,anti-inflammatory,and reduce apoptosis of egg cells by regulating targets such as AKT1,IL1 B,FOS,PTGS2,TNF,as well as cancer cell pathways,AGE-RAGE signaling pathways,interleukin-17 pathway and other related signaling pathways in diabetic complications. |
