Purpose:The tutor Professor Chen Min is a famous traditional Chinese medicine in Liaoning Province.The clinical application of traditional Chinese medicine in the treatment of senile insomnia has a good effect.Therefore,this study is a retrospective study.The method of data mining is used to study the traditional Chinese medicine prescription of Professor Chen Min in the clinical treatment of senile insomnia,so as to analyze and find the medication law of the tutor’s clinical application of traditional Chinese medicine in the treatment of senile insomnia,And explore the potential mechanism of its high-frequency drugs in the treatment of insomnia with the help of network pharmacology.Method:1.The elderly insomnia patients treated in Professor Chen Min’s outpatient department of the Affiliated Hospital of Liaoning University of traditional Chinese medicine from September 2020 to June 2021 were entered into the Microsoft Excel 2010 table.The ancient and modern medical case cloud platform software was used to analyze the frequency,sexual and flavor characteristics,association rules and cluster analysis of123 prescription drugs.Finally,the statistical results were analyzed and summarized by using the theory of traditional Chinese medicine,so as to summarize the medication law of Professor Chen Min in the treatment of senile insomnia.2.Carry out network pharmacology research on high-frequency drugs(Salvia miltiorrhiza,Glycyrrhiza uralensis,Schisandra chinensis,turmeric,white peony)obtained through data mining,screen the effective components and corresponding targets of high-frequency drugs in tcmsp database,screen disease targets in genecards database,and use Cytoscape 3 The network diagram of "high frequency drugs-effective chemical components-common target" was made by software 8.The protein interaction network(PPI)diagram was made by using string database,and the results were imported into Cytoscape 3 The key targets were screened in the software,and the common targets were analyzed by go analysis and KEGG enrichment analysis through metascape database to speculate the mechanism of high-frequency traditional Chinese medicine in the treatment of insomnia.Result:1.There were 123 elderly patients with insomnia,including 69 women and 54 men,aged from 60 to 70;There were 123 prescriptions for the first diagnosis,involving163 traditional Chinese medicines,including 34 high-frequency drugs with a frequency of ≥ 30 times.The top 15 drugs were Salvia miltiorrhiza,licorice,Schisandra chinensis,Yujin,white peony,Polygala tenuifolia,Poria,Prunella vulgaris,wild jujube kernel,corydalis Corydalis,keel,oyster,bupleurum,Ophiopogon japonicus and Pinellia ternata;The efficacy categories of high-frequency drugs are mainly tonic drugs,tranquilizing drugs,promoting blood circulation and removing blood stasis drugs and heat clearing drugs;The most frequently used drugs are mild and mild drugs;Sweet,pungent and bitter drugs are used more frequently;The drugs that are used more frequently for menstruation are drugs for liver and heart meridians;The drug pairs obtained by association rule analysis include Salvia miltiorrhiza Schisandra "," wild jujube kernel yanhusuo "," yanhusuo Danshen ",etc;Five clustering formulas were obtained by cluster analysis,class 1: Polygala tenuifolia,Poria,licorice,Salvia miltiorrhiza and Schisandra chinensis,class 2: Cinnamon Twig,Pinellia ternata,Scutellaria baicalensis,white peony,bupleurum,turmeric and Prunella vulgaris,class 3: Angelica sinensis,Paojiang,Phellodendron chinense and Coptis chinensis,class 4: Atractylodes macrocephala,Codonopsis pilosula,Fructus aurantii,Achyranthes bidentata,Polygonum multiflorum,Radix Pseudostellariae,wild jujube kernel,corydalis Corydalis,keel Oyster,raw land,Amomum villosum,Caulis Spatholobi,ginseng,cinnamon,cooked Rehmannia glutinosa.2.Network pharmacology research shows that there are 193 chemical components in high-frequency drugs(Salvia miltiorrhiza,licorice,Schisandra chinensis,Yujin and Paeonia alba),including 65 chemical components of Salvia miltiorrhiza,92 of licorice,8 of Schisandra chinensis,15 of Yujin and 13 of Paeonia alba.After weight removal,there are 187 chemical components and 255 action targets;1901 disease-related targets and 68 potential targets were obtained after intersection with high-frequency drugs;The components with higher degree value in the network diagram of "high frequency drugs-effective chemical components-common target" are quercetin,salvianol ketone β-Sitosterol,7-methoxy-2-methylisoflavone,betulinic acid.According to PPI network,the targets with high degree value are IL6,AKT1,TNF,IL1 B,VEGFA,CASP3,MMP9,FOS,PPARG and CXCL8;KEGG enrichment analysis obtained 286 pathways,including neuroactive receptor interaction,tumor necrosis factor(TNF)signal pathway,HIF-1 signal pathway and T-like receptor signal pathway.Conclusion:1.Tutor Professor Chen Min is good at treating senile insomnia with warm and mild drugs,sweet and pungent drugs;Starting from the whole,the five internal organs are in the same tune,mainly tonifying deficiency,supplemented by calming the mind,taking into account removing blood stasis and clearing heat.There are many symptoms of elderly patients.Professor Chen Min is good at grasping the main symptoms,screening out representative symptoms among many symptoms of elderly patients,selecting prescription and medication,combined with other symptoms,adding and subtracting with the syndrome and comprehensive conditioning,which can achieve good curative effect.2.High frequency drugs(Salvia miltiorrhiza,Glycyrrhiza uralensis,Schisandra chinensis,turmeric,white peony)may treat insomnia through quercetin,salvianol ketone β-Sitosterol,7-methoxy-2-methylisoflavone,betulinic acid and other active components act on targets such as IL6,AKT1,TNF,IL1 B and VEGFA,regulate the interaction of neuroactive ligand receptor and tumor necrosis factor(TNF)signal pathway,so as to provide reference for further research. |
