The research aims to explore the medication rules and academic thoughts of Professor Liu Longtao’s treatment of coronary heart disease based on the theory of blood stasis using data mining techniques and investigate the mechanism of action of the core prescription for coronary heart disease through network pharmacology and molecular docking technology.The first review discusses the application status of data mining in the core research areas of traditional Chinese medicine,such as the inheritance of experienced TCM doctors,clinical decision support systems,properties and efficiencies of Chinese herbal medicine,TCM diagnostic techniques,and TCM syndromes,from the perspectives of clinical research and theoretical research of traditional Chinese medicine.Literature review two comprehensively reviews the origin and relationship of blood-stasis-related concepts,summarizes the specific application of detoxification and activation of blood circulation method in coronary heart disease with blood stasis syndrome,and subsequently carries out the following work based on literature research:Objective:This research uses data mining techniques to deeply explore the implicit connotation behind Professor Liu Longtao’s medication rules for treating coronary heart disease based on the theory of blood stasis,formulating the core prescription for coronary heart disease with blood stasis syndrome,and predicting its potential mechanism of action through network pharmacology and molecular docking technology.Methods:1.The outpatient medical records of Professor Liu Longtao were screened based on inclusion/exclusion criteria and imported into Excel for data management.The data was uploaded to the Gujin Medical Cloud Platform V2.3.5 and the medication rules were mined using algorithms such as association rules,clustering analysis,and complex network analysis.The core prescription and academic thoughts were summarized using qualitative research methods.2.Target genes related to active components of drugs in the core prescription and coronary heart disease-related targets were screened from TCMSP and Genecards databases.Common targets of drugs and diseases were extracted,a PPI network and a drug-active component-action target network were constructed using Cytoscape,and GO and KEGG enrichment analyses were performed using Metascape and KEGG databases.Molecular docking technology was used to verify the binding degree between key targets and active components.Results1.The statistical analysis results show that the proportion of patients aged 60-69 years is the largest,and the number of visits in summer is significantly higher than other seasons,which reflects the age-related and seasonal characteristics of coronary heart disease.According to the meridian tropism of four qi and five flavors of traditional Chinese medicine,for the patients with blood stasis syndrome,the four qi are mainly warm and slightly cold,the five flavors are mainly sweet and bitter,and the meridian tropism is mainly heart,which embodies the core treatment principle of strengthening the body resistance and eliminating pathogenic factors.The analysis of TCM syndrome and treatment showed that Qi deficiency was the most common syndrome in patients with blood stasis and toxin syndrome,and the treatment of supplementing Qi,activating blood circulation and detoxifying was widely used in clinic.Syndrome-Chinese medicine association results show that the combination of Hedysarum Multijugum Maxim,Radix Salviae and Radix Paeoniae Rubra is widely used in patients with blood stasis syndrome with qi deficiency,phlegm turbidity and qi stagnation,which reflects the disease nature of coronary heart disease with blood stasis syndrome with deficiency in origin and excess in superficiality.Therefore,the method of supplementing qi and activating blood circulation has good universality in treating patients with qi deficiency or patients with phlegm turbidity and qi stagnation.The cluster analysis of traditional Chinese medicine showed that the top 20 commonly used traditional Chinese medicine could be divided into 4 groups and 8 sub-categories,which were specialized in supplementing qi and activating blood circulation,promoting qi and breaking blood,activating yang and dispersing masses,clearing away heat and toxic materials,calming nerves and relieving palpitation.Different drug combinations were adopted according to the clinical manifestations of patients with different pathogenesis of coronary heart disease.Complex network analysis of traditional Chinese medicine showed that the core prescription of traditional Chinese medicine was Hedysarum Multijugum Maxim,Glehniae Radix,Radix Salviae,Radix Paeoniae Rubra,Chuanxiong Rhizoma,Coptidis Rhizoma.2.The main targets of core prescription in treating coronary heart disease were AKT1,TP53,JUN,STAT3,etc.Through GO and KEGG enrichment analysis,it was found that the core prescription achieved the effect of treating coronary heart disease through various pathways and biological processes.The top three biological processes in GO enrichment pathway are the reaction of cells to nitrogen compounds,the reaction of cells to inorganic substances and the reaction of cells to organic cyclic compounds.The top three cellular components were membrane raft,transcription regulatory complex and receptor complex,and the top three molecular functions were DNA-binding transcription factor binding,protein-coupled amine receptor activity and protein domain-specific binding.The KEGG-rich pathways include cancer pathway,lipid and atherosclerosis,fluid shear stress and atherosclerosis,AGE-RAGE signaling pathway mediated diabetic complications,hepatitis B pathway,and PI3K-Akt signaling pathway.The molecular docking results showed that the top five binding energies from low to high were β-sitosterol and AKT1,bergapten and AKT 1,alloisoimperatorin and AKT1,quercetin and AKT1,quercetin and TP53,and their binding energies were-10.8 kcal/mol,-10.1 kcal/mol,-10 kcal/mol,-9.7 kcal/mol,-7.5 kcal/mol,respectively,all of which had good binding activity.Conclusion1.By analyzing the data,it was found that Professor Liu Longtao treated coronary heart disease syndrome of stasis and poison.According to the characteristics of the disease,he took Fuzheng and dispelling evil as the core treatment principle,and had the drug law of combining sweetness and bitterness,cold and warm.Combined with the qualitative interview method,the core prescriptions were determined to be Hedysarum Multijugum Maxim,Glehniae Radix,Radix Salviae,Radix Paeoniae Rubra,Chuanxiong Rhizoma,Coptidis Rhizoma.Its characteristic diagnosis and treatment ideas were based on the theory of stasis and toxin,combined with Chinese and Western ginseng,especially emphasizing tongue and pulse,emphasizing strengthening and protecting qi,flexible prescription,combination of tonifying and tonifying,and a wide range of medicinal flavors,reflecting the treatment characteristics of fine and hierarchical medication..2.The intervention of core formula on coronary heart disease has the characteristics of multi-component,multi-target and multi-pathway.It may be that quercetin,β-sitosterol,alloisoimperatorin,bergapten,cnidilin and other active components combine with common targets such as AKT1,TP53,JUN,STAT3 and participate in the regulation of cell death,inflammation,glucose metabolism,lipid metabolism and other related signal pathways and biological processes to finally achieve the effect of treating coronary heart disease. |
