Purpose: To use data mining technology to analyze the medication rules of Professor Qu Nini in the treatment of stable chronic obstructive pulmonary disease,summarize the tutor’s clinical experience in the treatment of this disease,and use network pharmacology technology to explore the mechanism of core prescriptions,in order to provide clinical treatment for the disease and new drug research and development to provide reference.Material and Methods: 1.Collect the medical records of patients with stable chronic obstructive pulmonary disease who were treated in the outpatient department of Professor Qu Nini,Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from September 2019 to September 2021.After screening according to the inclusion and exclusion criteria,they were sorted in an excel sheet,using Excel,IBM SPSS Modeler 18.0 and IBM SPSS Statistics 25.0 were used for frequency,association rules and cluster analysis.2.The effective active ingredients and action targets of the core prescriptions were screened out,and Cytoscape 3.8.0 was used to construct a "drug-ingredient-targets" network diagram,and the key active ingredients of the core prescriptions were obtained through network topology analysis;the disease targets of chronic obstructive pulmonary disease were obtained,The core drug action targets are obtained by intersecting the action targets of the core prescription and the disease targets.The protein interaction network is constructed through the String11.5 database,and then GO and KEGG enrichment analysis is performed through DAVID Bioinformatics Resources,and finally the core prescription is obtained.The key active components and mechanism of action of obstructive lung.Results:1.A total of 229 medical records were screened,among which there were 14 drugs with a frequency > 100 times,ranging from high to low: yam,tuckahoe,dogwood and so on.2.Perform association rule analysis(support is >50%,confidence is 100%),and finally 8 pairs of two herbs and medicines are obtained.It takes 58 pairs to get the three herbs.3.Cluster analysis was performed on high-frequency drugs,and they were divided into six categories: the first category(C1): Chuanxiong,Angelica;,Miziyuan;The third category(C3): Cortex Moutan,Alisma,Poria,Cornus,Schisandra,Chinese yam,Rehmannia glutinosa,Ophiopogon japonicus;The fourth category(C4): Astragalus,Codonopsis,Fried Chicken Neijin,North Sargassum;The fifth category(C5): Bupleurum chinensis,fried citrus aurantium with bran,and Fritillaria;the sixth category(C6): dried tangerine peel,fried Atractylodes with bran,Qing Pinellia.4.Using the network pharmacology research method,screened out the active ingredients of the core prescription traditional Chinese medicine: 20 Astragalus,2 Rehmannia glutinosa,16 Chinese yam,11 Cortex Moutan,10 Alisma,8 Schisandra,21 Codonopsis,Poria 15,20 dogwood,4 Ophiopogon japonicus.The top 10 active ingredients of core prescriptions obtained by network topology analysis.They are: quercetin,kaempferol,luteolin,7-O-methylisoxylitol,7-methoxy-2-methyl isoflavone,formononetin,beta-sitosterol,bean Sterols,isorhamnetin,3,9-di-O-methylnisolin.There are 167 key targets of core prescriptions for the treatment of stable COPD.The PPI network diagram of 167 key targets was constructed,and the top 10 core targets were obtained: TNF,IL1 B,JUN,CASP3,AKT1,PTGS2,VEGFA,EGFR,IL6,TP53,GO molecular biological process results: RNA polymerase II Positive regulation of promoter transcription,cytokine-mediated signaling pathway,positive regulation of gene expression,etc.;GO cell component results: cytosol,nucleus,plasma membrane,etc.;GO molecular function results: protein binding,same protein binding,enzyme binding,etc.;171 signaling pathways were obtained through KEGG analysis,mainly involving PI3K-Akt signaling pathway,MAPK signaling pathway,IL-17 signaling pathway,and TNF signaling pathway.Conclusion:1.The instructor recognizes that the main pathogenesis of chronic obstructive pulmonary disease in stable stage is the deficiency of lung and kidney qi,and the phlegm and blood stasis in the lung is the standard,and the deficiency is the main factor.Treatment should focus on nourishing the lungs and kidneys,absorbing qi and relieving asthma,taking into account the elimination of pathogenic factors,and always running through the idea of tonifying deficiency and reducing excess.2.Based on the results of network pharmacology,the core prescription has the characteristics of multi-component,multi-target and multi-pathway,and its mechanism of action may be to regulate AKT1,Targets such as JUN and EGFR play a role in regulating the release of inflammatory factors and the activation of inflammatory cells,improving airway remodeling,reducing airway mucus secretion,and delaying the progression of chronic obstructive pulmonary disease. |
