| Purpose:Based on the TCM Inheritance Computing Platform(V3.0),this study systematically excavates the medication rules of Professor Bai Guang for the treatment of functional diarrhea.Summarize the academic ideas of our teacher in the treatment of functional diarrhea,and deeply explore the connotation of the role between drug compatibility.The potential targets and mechanisms of action of core drug combinations in the treatment of functional diarrhea were explored through the method of network pharmacology.It provides more ideas and inspiration for the clinical treatment of functional diarrhea,and also provides a reference for basic research and new drug research and development.Material and method:Collected 129 patient cases with outpatient diagnosis of functional diarrhea diagnosed by Professor Bai Guang of the Affiliated Hospital of Liaoning University of Traditional Chinese Medicine from June 2019 to January 2022,and the first effective diagnosis.The collected patient case information and prescription drugs are entered into the Microsoft Office Excel(16.52)database.After the data is standardized,it is imported into the "Traditional Chinese Medicine Inheritance Calculation Platform(V3.0)" software for identification type distribution analysis and drug use law mining.The target of the core party is obtained from the TCMSP database,and the string11.5 database is introduced to build a protein interaction network.Use Cytoscape 3.8.0 to map the "Chinese Medicine-Active Ingredient-Target" network.Gene Ontology(GO)analysis of common targets of drugs and diseases was analyzed using R3.6.1 and the Kyoto Encyclopedia of Genes and Genomes(KEGG).Results:1.Based on the analysis of the basic information of the patients,it was found that among the 129 cases recorded in this study,there were 72 male cases,accounting for 55.81%,and 57 female cases,accounting for 44.19%.The oldest patients were 90 years old,the youngest 16 years old,and the mean age was 45.73 years old.Syndrome type analysis showed that the most patients were spleen and stomach weakness syndrome(37 cases),followed by spleen and kidney Yang deficiency syndrome(31 cases),cold and heat syndrome(30 cases),liver depression and spleen deficiency syndrome(18 cases),dampness-heat accumulation syndrome(13 cases).2.A total of 129 TCM prescriptions were included in this study,totaling 141 TCM prescriptions.According to the frequency analysis of Chinese medicines in the first diagnosis prescription,20 Chinese medicines with frequency≥30 were found,and the top five Chinese medicines were Tangerine peel,stir-fried Atractylodes macrocephala,stormwater,cinnamon and coptis chinensis.Analysis of the four qi,five flavor,meridian return and efficacy of the traditional Chinese medicine showed that warm medicine was the most frequently used,with 768 times of medicinal frequency.Among the five flavors,sweet taste had the highest frequency(955 times).The top 3 drugs were spleen meridian 1331 times,liver meridian 778 times,stomach meridian 769 times.The top three drug efficacy were tonifying deficiency 550 times,regulating qi 189 times and clearing heat 184 times.3.When the support degree was set as 20%and the confidence degree was set as 0.6,57 drug combinations were obtained.Among them,the top ten drug combinations were:①Tangerine peel and stir-fried Atractylodes atractylodes;②Tangerine peel,windproof;③fried Atractylodes macrocephalus,windbreak;④Tangerine peel and cinnamon;⑤Fried Atractylodes macrocephala and cinnamon;⑥Tangerine peel,coptis;⑦Fried Atractylodes atractylodes and Coptis;⑧Tangerine peel,peony root;⑨Orange peel,fried Atractylodes macrocephala,parsnip;⑩ Stir-fried Rhizoma atractylodes and prepared licorice.Using the"network display" function of TCM inheritance computing platform,the support degree was 20%,and the confidence degree was 0.6.The support degree was 50%,and the confidence degree was 0.9.Interdrug network diagram with 60%support and 0.9 confidence.According to the clustering algorithm of the complex system of the TCM inheritance assistance platform,25 new formulas were mined.4.Network pharmacologic analysis was carried out on the combinations of "Tangerine bark,Atractylodes macrocephalia and Rhizophora fangfeng" with support degree of 60%and confidence degree of 0.9.With OB(oral bioavailability)≥30%and DL(compound potency)≥0.18 as screening conditions,a total of 30 active components in core drug combinations"Orange bark,Atractylodes macrocephala and Nikip" acted on 91 drug targets.A total of 5628 disease targets for functional diarrhea were retrieved from GeneCards database.78 intersection targets were obtained,and GO and KEGG analyses were performed on the intersection targets.GO enrichment analysis results showed that the main responses to nutrition level,steroid hormone,lipopolysaccharide,oxidative stress,aging and other aspects play an important role.KEGG signal pathway enrichment analysis was conducted,and the results showed that "Tangerine bark,Atractylodes atractylodes and Fangfeng" may act on some factors in il-17 signal pathway,TNF signal pathway,endocrine resistance,apoptosis and other signal pathways,thus playing a role in the treatment of functional diarrhea.Conclusion:1.The distribution of the age of the disease collected in this study is mostly middle-aged,and the distribution of the patient’s symptoms is relatively average,and the most common symptoms are spleen and stomach weakness.2.Professor Bai Guang’s medication in the treatment of functional diarrhea is mainly tonic drugs,qi medicines,and antipyretic drugs.The analysis of the four qi showed that the frequency of use of warm medicines was the highest,the frequency of sweet taste in the five tastes was the highest,and the drugs were mostly attributed to the spleen meridian,the liver meridian and the stomach meridian.3.Based on the frequency of medication and cluster analysis of core combination,it was found that tongxie Yao Prescription was the basic formula for addition and subtraction in the treatment of functional diarrhea.In general,the basic principles were regulating the liver and spleen,invigorating the spleen and stopping diarrhea,and regulating cold and heat.4.The mechanism of action of the combination of "tangerine peel,white art,and parsnip"drugs in the treatment of functional diarrhea was predicted by using network pharmacology.It was found that drugs may play an important role by regulating responses to nutrient levels,responses to steroid hormones,responses to lipopolysaccharides,responses to oxidative stress,aging,etc.It may play a role in the treatment of functional diarrhea through certain factors in the signaling pathways such as IL-17 signaling pathway,TNF signaling pathway,endocrine resistance,and apoptosis. |
