Inflammation is the body’s defense response to external stimuli and is characterized by redness,swelling,fever,pain and dysfunction.Uncontrolled inflammation can lead to cancer,diabetes,stroke,chronic nephrosis,ischemic heart disease,non-alcoholic fatty liver,autoimmune and neurodegenerative diseases.Anti-inflammatory drugs have become the second large drug inferior to antibiotics.In the research of modern medicine,the molecular signaling pathways of the occurrence,development and transformation of inflammation have been explained,which providing a theoretical basis for the development of antiinflammatory drugs.China is rich in Chinese herbal medicines,and many Chinese herbal medicines have the effect of "clearing heat" or "removing fire".However,the parallel comparison between these medicinal materials is more ambiguous in traditional Chinese medicine.Based on the similarities in pathology between inflammation in modern medicine and "Shanghuo" in traditional Chinese medicine,our research team has established a technical platform for quantitatively evaluating the efficacy of drugs for reducing fire at the molecular level.This study used this platform to compare and screen the anti-inflammatory abilities of a group of 150 kinds of common traditional Chinese herbal medicines in parallel.The main results are as follows:(1)The crude components of 150 kinds of Chinese herbal medicines were extracted by ethanol condensation reflux method,and the ethanol extracts of 150 kinds of Chinese herbal medicines were obtained.(2)The protein level of the key immune protein Intercellular cell adhesion molecule-1(ICAM-1)in the inflammatory signaling pathway in Human microvascular endothelial cell(HMEC-1)was converted into inflammatory indexes.An inflammatory model induced by Lipopolysaccharides(LPS)was established,and the inflammatory index in the LPS group was defined as 100%.The model was used to screen 150 kinds of Chinese herbal medicines for anti-inflammatory effects in vitro.The 78 kinds of Chinese herbal medicines at the concentration of 512 μg/m L with the inflammation index lower than 30% were selected in the initial screening.Their toxic effects on HMEC-1 cells were further detected and 46 kinds of them were obtained that were not toxic to cells.There were 10 kinds of Chinese herbal medicines with the inflammatory index lower than 30% and 36 kinds of Chinese herbal medicines with the inflammation index above 30% when the concentration of the extract was reduced to 128 μg/m L in the re-screening.(3)Mice sepsis model was constructed,and the anti-inflammatory activity of Chrysanthemum indicum L.,Taraxacum mongolicum Hand.Mazz.,Agrimonia pilosa Ledeb.,Andrographis paniculate(Burm.f.)Nees,Peucedanum praeruptorum Dunn,Hedyotis diffusa Willd.,Alpinia oxyphylla Miq.,Bupleurum chinense DC.,Mentha haplocalyx Briq.,Artemisia argyi Levl.et Vant.with inflammatory index lower than 30%were verified in vitro.The expression of TNF-α,IL-1β and IL-6 in the serum of mice was detected by ELISA.The results showed that Hedyotis diffusa Willd.and Artemisia argyi Levl.et Vant.had the most significant inhibitory effect on the the expression of three inflammatory cytokines.Andrographis paniculate(Burm.f.)Nees and Chrysanthemum indicum L.had certain inhibitory effects on the expression of the three inflammatory cytokines.Peucedanum praeruptorum Dunn,Alpinia oxyphylla Miq.,Taraxacum mongolicum Hand.Mazz.and Mentha haplocalyx Briq.had significant inhibitory effects on the expression of TNF-α and IL-1β.These Chinese herbal medicines with high antiinflammatory effects can be applied to the development of anti-inflammatory active ingredients and the treatment of different inflammatory diseases in future. |
