Study On The Function Of Active Extract From Traditional Chinese Medicine: Evaluation On Inhibition To Metabolism-related Enzyme Activity And Anti-inflammation Effect

With the improvement of living standard and the development of science and technology,people pay more and more attention on natural, safe and innoxious Chinese herbal medicine plant. There also have many achievements which were obtained in research and development of Chinese herbal medicine plant. Green Tea, Lotus Leaf, Citrus Aurantum and Cassia Nomame have wide distribution and high production in China. Green tea is a type of tea made without fermentation.It contains more than 85% of polyphenols and theine.The functions of Lotus Leaf are reducing the blood fat; reduce weight, antibiosis and scavenging free radicals.The ingredients of Lotus Leaf are alkaloid flavonoid and essential oil.Citrus Aurantum contains alkaloid. It has the function of pressor effect, cardiotonic action, increase the flow volume of coronary and antisepsis.As a new plant phenolic compounds, Cassia nomame polyphenol can inhibit lipase obviously and prevent and treat obesity effectively, besides the common characteristic of most plant phenolic compounds.In this paper, we summarized the extracts of some Chinese herbal medicine plants, and the developing technologies and new methodology of lipase inhibition activity, a-glucosidase inhibition activity, anti-inflammatory and immunity. On the basis of it, with the research work of the lab, we did some research work for the extracts of Green tea, Lotus leaf, Citrus Aurantum and Cassia nomame,their polyphenol contentration, DPPH scavenging activity and the function of their lipase inhibition activity, a-glucosidase inhibition activity and anti-inflammatory activity. Finally, some significant results have been obtained.The main contents and results of this paper are as follows:(1)The extraction of the effective constituent from traditional Chinese medicine. Green Tea:80%Catechins and 50%EGCG,Lotus Leaf Extract:10:1,Citrus Aurantum: 10%Syneperine and Cassia Normame Leaf Extract:10:1.(2) The polyphenol contents of 4 kinds of Chinese herbal medicine plant were determined, using Folin-Ciocalteu method. The extract of Citrus Aurantum had highest polyphenol contents, which contains 462.5 mg/g. Lotus Leaf and Cassia Normame contain 293.8 mg/g and 270.3 mg/g.(3)Spectrophotometer method was used to analyses DPPH scavenging activity of 4 kinds of Chinese herbal medicine plant. The extract of Green Tea had highest DPPH scavenging activity. The second and third were Cassia Normame and Citrus Aurantum. IC50 are Green Tea:0.002 mg/ml,Cassia Normame:0.021 mg/ml,Lotus Leaf:0.037 mg/ml,Citrus Aurantum:0.061 mg/ml.(4) The lipase inhibition activities with Multilable Counter. The results showed that, with sequence as Cassia Normame>Citrus Aurantum>Lotus Leaf>Green Tea. IC50 are Cassia Normame:0.39 mg/ml, Citrus Aurantum:0.49 mg/ml, Lotus Leaf:0.59 mg/ml, Green Tea:1.40 mg/ml.(5)a-Glucosidase inhibition activities were assayed,using Spectrophotometer method.The results showed that, the extract of Green Tea had highest a-glucosidase inhibition activity. The second was Cassia Normame.Citrus Aurantum and Lotus Leaf were weaker comparatively. IC50 of Maltose group are Green Tea:0.01 mg/ml,Cassia Normame:0.40 mg/ml, Lotus Leaf:0.91 mg/ml, Citrus Aurantum:>8 mg/ml.Sucrose group are Green Tea:0.06 mg/ml, Cassia Normame:0.83 mg/ml,Lotus Leaf:0.97 mg/ml, Citrus Aurantum:>8 mg/ml.(6) The effect of anti-inflammatory activity of four kinds of herbs.The inhibitory effects of COX-2 and iNOS protein expression were performed in LPS-induced murine macrophages RAW264.7 cell,which is a model for inflammatory study. The results revealed that the extract of Cassia normame could effectively suppressed COX-2 and iNOS protein expression.
