The Study On The Mechanism Of Shenqi Guanxin Decoction Anti-atherosc Ierosis Based On Autophagy Theory

Objective: Under the guidance of the theory of "Benefiting qi and invigorating blood" in traditional Chinese medicine,the effects of Shenqi Guanxin Decoction on blood lipids,inflammatory indicators,aortic sclerosis,and expression of autophagy marker proteins in rats were experimentally explored to illustrate Shenqi Guanxin Decoction Whether it plays an anti-atherosclerotic effect by promoting macrophage autophagy and inhibiting inflammatory response,so as to provide experimental basis for the treatment of atherosclerosis and related diseases by traditional Chinese medicine and the theory of "Benefiting qi and invigorating blood " in traditional Chinese medicine.Methods: In this study,rat models of atherosclerosis with Qi-deficiency and blood-stasis syndrome were created by long-term cage feeding and long-term high-fat diet,which disturbed the sleep of rats.A total of 42 male SD rats with SPF grade were selected in the experiment,and they were randomly divided into 6 groups,7 rats in each group,divided into control group,model group,simvastatin group,"Shenqi Guanxin Decoction" high-dose group(SQGX-H),medium dose group(SQGX-M),low dose group(SQGX-L),after 7 days of adaptive feeding,start the test,except the control group,the other groups are fed with high-fat feed(high-fat feed formula: 2 % cholesterol + 0.5% sodium cholate + 3% lard +0.2% propylthiouracil + 94.3% + vitamin D3 powder(1.25x106IU/kg),use different light time to interfere with sleep,after feeding for 8w,the establishment of Qi deficiency and blood stasis syndrome Rat atherosclerosis model.After the model was established,the simvastatin group(5mg/kg/d),the "Shenqi Guanxin Decoction" high-dose group(40g/kg/d),the middle-dose group(20g/kg/d)),low-dose group(10g/kg/d),each group was intervened for 4 weeks,and then the expression of blood lipids,inflammatory indicators,and autophagy marker proteins were detected,and the lipid status of aortic plaque was observed to study whether Shenqi Guanxin Decoction is By promoting macrophage autophagy,affecting lipid metabolism and inhibiting vascular inflammatory response,it exerts an anti-atherosclerotic effect.Results: The experiment showed that the medium and high dose groups of Shenqi Guanxin Decoction had the effect of significantly reducing TC,TG,and LDL-C in rat serum(P<0.01)compared with the model group(P<0.01),and could increase HDL-C(P<0.05),at the same time,it can significantly reduce IL-1β and IL-6 inflammatory indicators(P<0.01).Oil red O staining suggests that the high-dose Shenqi Guanxin Decoction group can improve atherosclerosis.Stability,the expression of autophagy-related proteins LC3-Ⅱ,LC3-Ⅰ,LC3-Ⅱ/LC3-Ⅰ and p62 were detected by Western blot.It can be seen that the expression of LC3-Ⅱ protein was significantly increased,and the ratio of LC3-Ⅱ/LC3-Ⅰ was significantly increased,with a very significant statistical difference(P<0.001).decreased,with a highly significant statistical difference(P<0.001).Conclusions:1.The middle dose group(crude drug 20g/kg/d)and high dose group(crude drug 40g/kg/d)of Shenqi Guanxin Decoction can significantly reduce the levels of TG,TC,LDL-C in the serum of AS model rats and Raise HDL-C levels.2.The mid-dose group(crude drug 20g/kg/d)and high-dose group(crude drug 40g/kg/d)of Shenqi Guanxin Decoction can reduce the inflammatory indexes of IL-1β and IL-6 in AS model rats.3.The high-dose group of Shenqi Guanxin Decoction(crude drug 40g/kg/d)can improve atherosclerosis in AS model rats.4.The high-dose group of Shenqi Guanxin Decoction(crude drug 40g/kg/d)can increase the expression of macrophage autophagy-related proteins LC3 Ⅱ and LC3Ⅱ/Ⅰ in the aortic tissue of AS model rats,and reduce the expression of p62,It shows that Shenqi Guanxin Decoction can enhance autophagy activity and play an anti-AS role.In summary,the anti-AS mechanism of Shenqi Guanxin Decoction may be through regulating the autophagy of macrophages in arterial tissue and affecting lipid metabolism.
