Research background and purpose:Osteoarthritis(OA)is a chronic joint disease with a high incidence rate among middle-aged and elderly people.The classic Chinese medicine prescription"Hai Tong Pi Decoction"is often used to treat OA by fumigation and washing,with a clear therapeutic effect.However,the fumigation and washing method is fussy and time-consuming,the temperature of the liquid medicine is not easy to control,there is no uniform standard for the concentration of the liquid medicine,and the efficacy of the medicine can’tbe guaranteed.In order to simplify the operation,prolong the administration time,reduce the times of administration and maintain the effective therapeutic concentration,it is necessary to develop the traditional decoction into a modern external Chinese medicine preparation.There are twelve taste of drugs in the original prescription of"Hai Tong Pi Decoction",so it is difficult to prepare it.Therefore,in this study,the taste of the medicine was reduced and it was cut into"Modified Hai Tong Pi Decoction".In order to ensure its key quality attributes and lay a foundation for the next step of preparing qualified compound preparations of traditional Chinese medicine,this study carried out a preliminary study on the medicinal properties.Materials and methods:1.Based on the theory of prescription compatibility,the original prescription was cut into Modified Hai Tong Pi Decoction,and the rationality and effectiveness of Modified Hai Tong Pi Decoction were preliminarily verified by cell pharmacodynamics experiments.2.Exploring the material basis and mechanism of Modified Hai Tong Pi Decoction in treating OA by means of network pharmacology,and screening the Indicator components of each medicine in the prescription by UPLC-Q-TOF-MS/MS technology.3.The extraction technology of Erythrina bark,Impatiens balsamina,Olibanum and Myrrha were studied respectively,and the drying technology of the extracts was explored.Results:1.The prescription of the Modified Hai Tong Pi Decoction was determined as follows:6 g of Erythrina bark,6 g of Impatiens Balsamina L,6 g of frankincense and 6g of myrrh.The optimal inflammatory concentration of LPS was 2 ng/m L,and the inflammatory model of rat chondrocytes was successfully established.Taking cell morphology,cell survival rate and inflammatory factors as indexes,it is determined that the serum containing 10%Modified Hai Tong Pi Decoctioncan effectively intervene chondrocyte inflammation.2.With the help of network pharmacology and UPLC-Q-TOF-MS/MS technology,four measurable effective components including Erythraline,Erysotrine,erysodienone,and Erysodinewere screened from Erythrina bark;Three measurable effective components namely quercetin,kaempferol and scopolamine were screened from Impatiensbalsamina L;Selected from frankincensewere three measurable active ingredients includingβ-Elemenone acid,α-Mastic acid andβ-Mastic acid;Unable to screen out measurable active ingredients from myrrhwithout clear drug identification.3.The acid dye colorimetric method was used to investigate the total alkaloid content,and the optimal extraction process for Erythrina bark was obtained by adding 7times the amount of 82%ethanol,extracting twice for 78 minutes each time;The content of quercetin,kaempferol,and scopolamine was investigated by HPLC,and the optimal extraction process for Impatiens balsamina L was obtained by adding 5 times the amount of 82%ethanol and reflux extraction for 35 minutes;Using HPLC method forβ-Elemenone acid,α-Mastic acid andβ-Mastic acid.The content of mastic acid was investigated and the optimal extraction process for mastic was obtained by adding 8times the amount of 92%ethanol and extracting twice with ultrasound for 87 minutes each time;Taking the unit total peak area at a wavelength of 300 nm as the indicator,the optimal extraction process for myrrh was obtained by adding 16 times the amount of96%ethanol and extracting twice with ultrasound for 52 minutes each time.The best spray drying process of the Modified Hai Tong Pi Decoctionextract mixed according to the prescription amount is that the alcohol extract with relative density of 0.90 g/cm~3is used as the material,the air inlet pressure is 0.5 MPa,the feed rate is 15m L/min,and the air inlet temperature is 130℃.Conclusions:Based on the theory of prescription compatibility,this study analyzed the rationality of Hai Tong Pi Decoction,and preliminarily discussed its effectiveness through cell pharmacodynamic experiments.With the help of network pharmacology and LC-MS technology,the quality inspection indexes of this prescription for OA were screened out,and the best extraction process and drying process of each drug flavor were optimized,which laid the foundation for the next preparation research. |
