Background: In recent years,the medicinal and food homologous plant polygonatum has become a research hotspot.The polygonatum can be used only after processing.Nine steaming and nine drying are the most widely used and the oldest method with the effect of reducing toxicity and increasing efficiency.Modern research showed that different processing times in the process of "nine steaming and nine drying" will cause differences in the component of polygonatum,which may lead to changes in the metabolites of organisms after taking polygonatum.Metabolomics can be used as a tool to reveal the metabolic fluctuations in the body caused by different processing times of polygonatum.Purpose: Analyze the nutritional and chemical components of raw polygonatum,two steaming and two dryingand polygonatum,nine steaming and nine drying polygonatum,and reveal the effects of three groups of polygonatum on organism metabolism under different administration time through plasma metabolomics,and finally find the relationship between active component and metabolites through network connection.Experiment design: According to the method specified in the standard of the American Association of Cereal Chemists,the nutritional components of raw polygonatum,two steaming and two dryingand polygonatum,nine steaming and nine drying polygonatum were analyzed.At the same time,UPLC-QTOF/MS and UNIFI were used to analyze the chemical components of three groups of polygonatum,and Mass Lynx was used to analyze the content.Results: 1.There are significant differences in the contents of protein,fat and carbohydrate in the nutritional components except ash.2.There are 29 chemical components in raw polygonatum,26 chemical components in two steaming and two dryingand polygonatum,31 chemical components in nine steaming and nine drying polygonatum,and most of them have significant differences.3.The common pathways of 5-day and 13-day administration are glycerol phospholipid metabolism and primary bile acid biosynthesis.In addition,both have different metabolic pathways.4.The two active components of 4’,5-dihydroxyflavone and liquiritigenin affect the glycerol phospholipid metabolism pathway and promote the metabolism of glycerol phospholipids by activating PLA2 enzyme.Conclusion: There are not only differences in nutritional components,but also differences in chemical components and content of raw polygonatum,two steaming and two drying polygonatum,nine steaming and nine drying polygonatum.After administration of three groups of polygonatum,the metabolism of organism will change,and the changes will also be different with the increase of administration time.4’,5-dihydroxyflavone and liquiritigenin play a positive role in promoting the metabolism of glycerol phospholipids,and the effect of nine steaming and nine drying polygonatum is stronger than that of the other two groups in 13 days of administration.
