B.scorzonerifolium Wil Id.is a perennial herb of the genus Bupleurum L.and Umbelliferae.B.scorzonerifolium Wil Id.has received close attention in recent years because of its very important development and utilization value in the field of medicine.In this thesis,the chemical constituents of B.scorzonerifolium Wil Id.were studied,and 14compounds were isolated.The anti-hepatocarcinoma mechanism of saikosaponins was successfully analyzed by using the research method of network pharmacology for the first time,and the accuracy of the prediction results was verified by cytotoxic activity assay.In this thesis,14 compounds were isolated from B.scorzonerifolium Wil Id.by macroporous resin column chromatography,silica gel column chromatography and HPLC and so on.The structures of all compounds were identified by physical and chemical properties and NMR and HRESIMS spectroscopy,including 10 saponins,1 monoterpene,1 lignin and 1 organic acid compounds.These compounds were i dentified as:Saikogenin F(1),Saikogenin G(2),3β,16β,23,28-tetrahydroxy-11α-methoxy-olean-12-ene-3-O-β-D-fucopyranoside(3),6’’-O-acetylsaikosaponin a(4),6’’-O-acetylsaikosaponin b4(5),6’’-O-acetylsaikosaponin d(6),Saikosaponin b2(7),Sai kosaponin b3(8),Saikosaponin b4(9),3β,23,28-trihydroxy-11α-methoxy-12-ene-16-one-3-O-β-D-glucopyranosyl-(1→3)-β-D-fucopyranoside(10),Syringaresinol 4’-O-β-D-glucopyranoside(11),(2R,3R)-2β-(4’’-hydroxy-3’’-methoxybenzyl)-3α-(4’-hydroxy-3’-m ethoxybenzyl)-γ-butyrolactone 2-O-(β-D-glucopyranoside)(12),Bornyl 6-O-β-D-apiofu ranosyl(1→6)-β-D-glucopyranoside(13)and Methyl-9,12,13-trihydroxyoctadeca-10,15-dienoate(14),compound 10 is a new compound that has not been reported in the literature.The components,target proteins and mechanism of saikosaponins against hepatocellular carcinoma(HCC)were analyzed by using the method of network pharmacology.Through the network analysis,a total of 6 saikosaponins were screened as active components,6 key target proteins were screened from 40 target proteins related hepatocarcinoma,and the genes related to potential targets were analyzed by GEPIA,GO,KEGG enrichment analysis.The mechanism of multi-component and multi-target action of saikosaponins and its effect on its prognosis were explained.Then the cytotoxic activity of the isolated saikosaponins 1-10 was tested based on MTT method..The results showed that compound 4、6、7、8 showed obvious cytotoxic activity.
