Objectives:1.Using data mining technology to analyze the treatment ideas of Professor Jingri Xie in treating patients with hepatitis B cirrhosis ascites with blood stasis and water stagnation syndrome,and summarize Professor Jingri Xie’s medication experience and core prescriptions.2.To investigate the potential targets and key pathways of action of core prescriptions for the treatment of hepatitis B cirrhosis ascites using a network pharmacology approach,and to provide a preliminary basic theoretical basis for subsequent experimental studies.Methods:1.The clinical data of patients with hepatitis B cirrhosis with ascites with blood stasis and water stagnation syndrome were collected from June 2011 to June 2021 who attended the clinic of Professor Jingri Xie at the First Affiliated Hospital of Heilongjiang University of Traditional Chinese Medicine.Data mining is performed on patients’ general conditions,four diagnostic information and herbal prescription data to analyze the basic frequency situation,then grouping common drug pair combinations based on association rules and exploring drug clusters using systematic clustering,and finally constructing core prescriptions through core networks.2.The TCMSP data platform was used to find and filter the active ingredients and corresponding targets of the core prescriptions,and the targets were standardized according to the UniProt platform data.In the "GeneCards","OMIM" and "DurgBank" databases,the keywords "Liver cirrhosis ascites","Cirrhotic ascites" and "Liver ascites" were searched to obtain disease-related targets,and the targets were intersected with targets of core prescriptions to obtain drug-disease intersection targets.The intersection targets were analyzed by STRING data platform,PPI network maps were drawn,and the platform data were imported into CytoScape3.9.1 to filter out the core targets.The intersection targets were imported into the DAVID data platform for GO and KEGG enrichment analysis.Results:1.A total of 174 cases were included in this study,including 109 males and 6 5 females,with ages ranging from 40 to 70 years.In descending order of frequency,the top 3 clinical manifestations were dorsal rib stabbing pain,abdominal bulging,and poor appetite;the 1st ranked tongue sign was petechiae on the tongue;and the 1st ranked pulse sign was astringent pulse.2.A total of 174 prescriptions containing 124 flavors were entered,of which the majority of the medicinal properties were warm,followed by plain and slightly cold;most of the tastes were pungent,followed by sweet and bitter;most of the meridians were liver and spleen meridians;and the top ten ranked in frequency were Angelicae sinensis radix,Chuanxiong hizome,Poria,Atractylodis macrocephalae rhizoma,Astragali radix,Alismatis rhizoma,Lycopi herba,Magnoliae officinalis cortex and Andcurcumae rhizoma.3.A total of 51 association rules were obtained,and 7 common drug pairs were screened out.A total of 7 combinations were obtained by systematic clustering,which were divided into three groups:"activating blood circulation method cluster","water-relieving method cluster" and "regulating Qi-flowing method cluster".The core network was constructed and the core prescriptions were obtained as:Angelicae sinensis radix,Chuanxiong hizome,Poria,Atractylodis macrocephalae rhizoma,Astragali radix,Alismatis rhizome and Lycopi herb a4.The core prescription drugs were searched through the TCMSP platform,and 57 active active ingredients were screened,corresponding to 26 5 targets.The disease database obtained 924 targets,and a total of 98 drug-disease intersection targets were obtained by taking intersection with drug targets,and 20 core targets were obtained after filtering.GO analysis suggests that the core prescriptions are likely to involve binding to proteins,transcription factors,and characteristic enzymes involved in gene expression.KEGG analysis speculates that the core prescriptions are more closely related to processes such as cancer,viral infection,inflammation,and oxidative stress.Conclusions:1.Prof.Jingri Xie treats hepatitis B cirrhosis ascites with blood stasis and water stagnation as the basic treatment method throughout the treatment,combine with products that promoting Qi and resolving dampness,paying attention to the hierarchy of blood stasis and the addition and subtraction of drugs according to the changes of the disease mechanism with the individual’s constitution,and the treatment of water stagnation starts from regulating the water channels of the internal organs and treats from the deficiency of Qi and blood.2.The core prescription of Prof.Jingri Xie for the treatment of hepatitis B cirrhosis ascites with blood stasis and water stagnation has multi-component,multi-target and overall regulatory action characteristics,mainly involved in anti-inflammatory,antioxidant,antiviral infection and other mechanisms of action.
