Objective:To analyze the prescriptions of Professor Li Haisong for treating patients with chronic prostatitis of qi stagnation and blood stasis through the use of data mining on the auxiliary platform of traditional Chinese medicine inheritance,and to explore the rules of medication,core prescriptions and core Chinese medicines.Then conduct network pharmacology analysis of core drugs to explore the targets,signal pathways and mechanism of action of core drugs in the treatment of chronic prostatitis.Lay the foundation for the screening and experimental verification of traditional Chinese medicine for the treatment of chronic prostatitis with stagnation of qi and blood.Methods:(1)Collect and sort out Professor Li Haisong’s prescriptions for patients with chronic prostatitis of Qi stagnation and blood stasis type from September 2018 to June 2019.Through the traditional Chinese medicine inheritance assistance platform software(TCMISS),the prescription of the medicine’s nature and taste,the frequency of use,etc.are analyzed;based on the association rules,the commonly used drug combinations are obtained;then the core drugs and prescriptions are obtained;the new analysis is based on the complex system entropy clustering method prescription.(2)Retrieve the effective active ingredients of the former core drugs through the TCM database(TCMSP,http://tcmspw.com/tcmsp.php),and use the Uniprot(https://www.uniprot.org/)database to retrieve the The target protein name is converted to the target abbreviation for standardization.Search chronic prostate(search term:prostatitis)through Genecards database(https://www.genecards.org/)to obtain disease targets of chronic prostatitis.The target of the active ingredient of the core prescription drug and the target of the disease are intersected to obtain the active ingredient and potential target.Construct a protein-protein interaction(PPI)network model on the STRING platform(https://string-db.org/)with the common targets obtained through screening.Import the data into Cytoscape 3.8.1 software to analyze the network topology and screen out the core targets.Import the core target into the DAVID data analysis platform for GO and KEGG enrichment analysis.Results:(1)Prescription analysis part:A total of 326 prescriptions were included,involving 145 flavors of drugs.The proportion of medicinal properties from high to low is warm,cold,calm,cool,and hot;the medicinal taste in descending order of proportion is bitter,pungent,sweet,sour,salty,and astringent;the meridian in descending order of proportion is liver,spleen,Stomach,lung,heart,kidney,gallbladder,large intestine,bladder,pericardium,small intestine,triple burnt.According to the analysis of association rules and complex system entropy clustering method,6 groups of core drug combinations and new prescriptions are obtained.The core prescription consists of the top 10 drugs:Baishao,Astragalus,Poria,Bupleurum,Wangbuliuxing,Danshen,Qingpi,Angelica,Zhigancao,Corydalis.(2)Network pharmacology analysis part:The drugs in the core prescription act on chronic prostatitis in a multi-component,multi-target,and multi-channel way.Results There are 249 intersection targets between the color core prescription and chronic prostatitis.By using string database analysis to construct a protein interaction network,there are 4447 interactions among 249 targets.The data was imported into Cytoscape 3.8.1 software for network topology analysis and related screening,finally 44 core targets were obtained.The core target is closely related to the inflammation and immune response during the pathogenesis of chronic prostatitis.The functions of GO enrichment analysis are mainly related to gene expression,protein binding and other biological processes and molecular functions,and are related to inflammation and cancer progression.Among the top 20 pathways with a high degree of KEGG enrichment,they are closely related to cancer response,inflammatory response,and oxidative stress.Conclusion:Through data mining,Professor Li Haisong’s core prescription for the treatment of chronic prostatitis with qi stagnation and blood stasis by promoting qi and blood stasis is composed of white peony root,astragalus,Poria,Bupleurum,Wangbuliuxing,salvia,green peel,angelica,and licorice,Yanhusuo constitutes.Based on the method of network pharmacology,the core prescriptions for the treatment of chronic prostatitis of Qi stagnation and blood stasis type are quercetin,kaempferol,stigmasterol,isorhamnetin and so on.The core targets are PTGS2,HSP90AB1,PTGS1,SCN5A and other targets.Through GO analysis,it is found that the core prescription has biological processes and molecular functions such as regulating gene expression and protein binding.KEGG pathway analysis results indicate that cancer pathways,prostate cancer,TNF signaling pathways,FoxO signaling pathways,estrogen signaling pathways,etc.may be the main pathways through which the core prescription plays a role. |
