Objective:1.Application of data mining to summarize the medication rules and core drug and explore the core prescriptions for the treatment of spleen deficiency of ovarian abnormal uterine bleeding in ancient books of gynecology,explore the medication experience of ancient doctors in the treatment of spleen deficiency of ovarian abnormal uterine bleeding,and provide theoretical basis for clinical practice.2.Conducting network pharmacological to analysis core drugs obtained from data mining,preliminarily explore the mechanism of action of core drugs in the treatment of spleen deficiency of ovarian abnormal uterine bleeding,screen out the active components,predict their potential targets and related pathways,and provide reference for clinical practice.Methods:1.The prescriptions for the treatment of spleen deficiency of ovarian abnormal uterine bleeding in the Integrated Classic Works of Gynecology of Traditional Chinese Medicine were screened out,and the names of the herbs were standardized and entered into the Excel table to establish the database of spleen deficiency of ovarian abnormal uterine bleeding.The "Traditional Chinese Medicine Inheritance Computing Platform 3.0" was used for statistical analysis,and the frequency of drug,association rules and cluster analysis results were obtained.To explore the medication rules,core drugs and core prescriptions for spleen deficiency of ovarian abnormal uterine bleeding in ancient gynecological books of traditional Chinese medicine.2.Network pharmacology analysis of core drugs obtained by data mining was carried out,the active components of core drugs were obtained through TCMSP and their possible targets were predicted,the predicted targets were normalized by Uni Prot database.The "drug-component-target" network was constructed by Cytoscape 3.9.1.Drug Bank、 OMIM and Gene Cards databases were used to obtain the disease targets related to ovarian abnormal uterine bleeding.The online program Venny 2.1.0 was used to obtain the intersection targets of drugs and diseases,and the intersection targets were uploaded to the STRING platform to obtain the protein-protein interaction network.Cytoscape 3.9.1 was used for visual analysis.The Metascape platform was used to perform GO functional enrichment analysis and KEGG pathway enrichment analysis of intersection targets.To explore the mechanism of core drugs in the treatment of spleen deficiency of ovarian abnormal uterine bleeding by screening the active components of core drugs,predicting their potential targets and related signaling pathways.Results:1.In this study,77 prescriptions,involving 81 Chinese herbs for the treatment of spleen deficiency of ovarian abnormal uterine bleeding in ancient gynecological works of Traditional Chinese Medicine were obtained.The cumulative frequency of medication was 779 times,6 kinds of drugs were used more than 30 times,which were Atractylodes,Licorice,Angelica sinensis,Ginseng,Astragalus membranaceus and Radix paeoniae alba.The frequency of tonic drugs were significantly higher than other efficacy drugs,up to 405 times,accounting for 52.25%.Warm drugs were the most frequent occurrence of four qi of the drug,437 times,accounting for 56.10%,followed by cold,flat,hot and benign drugs.Sweet medicines were used 485 times,accounting for 41.88%,followed by bitter,pungent,sour and salty.All 12 meridians were involved,mainly the five internal organs,and the spleen meridian was the most,538 times,accounting for 24.86%.A total of 44 medication patterns and 79 association rules were obtained by drug association rule analysis.There were 10 core drugs: Atractylodes macrocephala,Licorice,Angelica sinensis,Astragalus membranaceus,ginseng,Rehmanniae radix,Poria cocos,Radix paeoniae radix,Radix bupleurum and Tangerine peel.Three core prescriptions were obtained by drug cluster analysis,core prescription 1: Angelica sinensis,Atractylodes macrocephala,Radix paeoniae alba,Prepared rehmannia root,Ligusticum wallichii;core prescription 2: Atractylodes macrocephala,Licorice,Ginseng,Angelica sinensis,Cuttlebone;core prescription 3: Atractylodes macrocephala,Licorice,Ginseng,Angelica sinensis,Astragalus membranaceus.2.A total of 135 active ingredients and 206 potential targets of core drugs were obtained through network pharmacology.In the "drug-ingredient-target" network,the node degree value of quercetin was significantly higher than that of other active ingredients.A total of 1744 disease targets were obtained.There were 119 core drug and disease intersection targets.The core targets were mainly AKT1,TP53,IL6,TNF,and VEGFA.The core drugs involved 1630 biological processes,75 cellular components,141 molecular functions and 192 signaling pathways,mainly involving PI3K-AKT signaling pathway,TNF signaling pathway and NF-κB signaling pathway.Conclusions:1.The prescriptions,included in this study,for the treatment of spleen deficiency of ovarian abnormal uterine bleeding recorded in ancient gynecological books of traditional Chinese medicine are mainly tonic drugs,sweet and warm drugs,mainly returning to the spleen meridian.2.The core medicines for the treatment of spleen deficiency of ovarian abnormal uterine bleeding by ancient Chinese medicine are Atractylodes macrocephala,Licorice,Angelica sinensis,Astragalus membranaceus,ginseng,Rehmanniae radix,Poria cocos,Radix paeoniae radix,Radix bupleurum and Tangerine peel.Three core prescriptions were obtained by cluster analysis,core prescription 1: Angelica sinensis,Atractylodes macrocephala,Radix paeoniae alba,Prepared rehmannia root,Ligusticum wallichii;core prescription 2: Atractylodes macrocephala,Licorice,Ginseng,Angelica sinensis,Cuttlebone;core prescription 3: Atractylodes macrocephala,Licorice,Ginseng,Angelica sinensis,Astragalus membranaceus.3.The main active ingredient of the core drug are quercetin,kaempferol,masterol and β-sitosterol,which may act on AKT1,TP53,IL6,TNF,VEGFA and other targets in terms of biological functions such as response to hormone,protein kinase activity and cytokine activity,regulate PI3K-AKT signaling pathway,TNF signaling pathway,NF-κB signaling pathway and other pathways,to inhibit inflammatory response,promote platelet aggregation and endometrial angiogenesis,stimulate follicle development,restore ovulation,and thus adjust menstrual cycle to treat spleen deficiency of ovarian abnormal uterine bleeding.
