A Study On The Medication Rules Of Traditional Chinese Medicine For Adenomyosis Based On Data Mining And Network Pharmacology

Objective:1.The medical records of the real world clinical treatment of adenomyosis with traditional Chinese medicine were extracted,and the law of high-frequency drugs and core prescriptions of traditional Chinese medicine in the treatment of adenomyosis was summarized by using the method of data mining.2.According to the core prescription obtained from data mining,the mechanism of network pharmacology in the treatment of adenomyosis was discussed from multiple levels and multiple angles.Method:This study is divided into two parts.The first part is data mining.The information of patients who visited the gynecological outpatient department and ward of the affiliated hospital of shandong university of traditional Chinese medicine from 2012 to 2017 was extracted,and those diagnosed as adenomyosis by gynecological sonography,with complete basic information and treated with traditional Chinese medicine or single herbal concentrate-granules were included in the database.Frequency analysis and the complex network software Liquorice system in the data warehouse platform were used to mine and analyze the medication rules of adenomyosis.The second part is network pharmacology.The core of the prescription of the data mining in TCMSP database(http://tcmspw.com/tcmsp.php)for Chinese traditional medicine compound component retrieval and target forecast,in Genecards database(https://www.genecards.org/)for Adenomyosis associated genetic information retrieval,using Venn diagram targets mapping,through the String database(https://string-db.org/),Protein-protein interaction network diagram,enriched with David database(https://davidncifcrf.gov/)for analysis of reuse Omicshare website(https://www.omicshare.com/)to analysis the results visualization.Results:1.A total of 8986 prescriptions were collected.362 kinds of traditional Chinese medicines were used.The frequency of using vinegar was the highest,followed by Angelica,Chuanxiong,vinegar Yanhusuo and Puhuang.The frequency of using them was more than 5000.The medicine was mainly warm and flat.The taste of medicine was mainly bitter,sweet and pungent.The meridians were mainly liver and spleen meridians.The efficacy was mainly promoting blood circulation and eliminating stasis and tonifying deficiency.The sub network diagram of the core prescription of Angelica,vinegar Xiangfu,Chuanxiong,Puhuang,vinegar Yanhusuo,white peony,Dangshen and three layers of core drugs addition and subtraction was obtained.2.By searching the core prescription,101 compounds were obtained,174 predicted targets and 155 gene names could be transformed into human genes.There are 27 targets in the core prescription.284 genes related to"adenomyosis" were retrieved and mapped with 155 gene names in the core prescription.32 common gene names were obtained.After enrichment analysis,31 items related to biological process,3 items related to cell composition,19 items related to molecular function and 48 signal pathways were obtained.Conclusion:1.The treatment of adenomyosis should be based on the general premise of promoting blood circulation and removing blood stasis,with the help of regulating qi and relieving pain,supplementing qi and replenishing blood.According to the clinical symptoms of the patients,the drugs can be added or subtracted.Those with cold coagulation and blood stasis can warm the meridians and disperse the cold,those with hot burning and blood stasis can clear away the heat and cool the blood and remove the blood stasis,those with qi stagnation and blood stasis can activate Qi,activate blood circulation and relieve pain,those with weak vital energy can nourish Qi and blood,and those with kidney deficiency and blood stasis can replenish kidney and fill essence.2.The core prescription treatment of adenomyosis involves several key targets such as VEGFA,MMP2,ESR1,ESR2,PGR,PTGS2,and signal pathways such as HIF-1,TNF,PI3K Akt,NF-κB.The treatment of adenomyosis with multiple approaches and targets embodies the characteristics of treating diseases with multiple drugs,multiple components and multiple targets in traditional Chinese medicine compound.
