Discussion Of Professor Li Qiuyan’s Medication Rules And Core Prescription Mechanism In The Treatment Of Climacteric Syndrome

Climacteric syndrome is a series of physical and mental symptoms caused by the fluctuation or decrease of hormone levels before and after menopause,and is one of the common chronic diseases of women.In the process of the aging of the world population,the incidence rate of climacteric syndrome is also gradually increasing,which has caused serious physical and mental impact on women.Traditional Chinese Medicine pays attention to holistic therapy,which can ameliorate the idiosoma and mentality symptoms of patients with climacteric syndrome at the same time,and has its unique advantages in treatment.At present,the treatment of climacteric syndrome with traditional Chinese medicine is one of the hot points in the educational circles.Professor Li Qiuyan has rich experience in diagnosis and treatment of climacteric syndrome,following the guidance of Weng Weiliang,a master of traditional Chinese medicine.This study uses data mining technology to summarize Professor Li Qiuyan’s medication rules in the treatment of climacteric syndrome,and excavate the core prescription to explore Professor Li Qiuyan’s academic thoughts.Based on the network pharmacology and molecular docking technology,the potential mechanism of the core prescription in the treatment of climacteric syndrome was discussed to lay a foundation for the follow-up basic research.Study 1:Analysis of Professor Li Qiuyan’s medication law in the treatment of climacteric syndromeObjective:Through data mining method,to extract the medication rules of Professor Li Qiuyan in the treatment of climacteric syndrome and to explore the core prescription.Methods:By sorting out the outpatient medical records of Professor Li Qiuyan in the treatment of climacteric syndrome for nearly 10 years(June 2012 to June 2022),the medical records were summarized and standardized,and the original database was established by Excel.The data were imported into SPSS Modeler,SPSS Statistics and Gephi respectively for frequency statistics,association rule analysis,cluster analysis and complex network analysis of the four gases,five flavors and meridian tropism of traditional Chinese medicine.Analyze Professor Li Qiuyan’s medication frequency,drug compatibility and core prescription drug combination,and summarize Professor Li Qiuyan’s medication rule and treatment experience.Results:242 patients and 574 prescriptions of traditional Chinese medicine were included in this study.All patients were women,age-bracket from 44 to 58 years old.Among them,patients in the age range of 48 to 53 were the most,accounting for 76.86%of the total number.A total of 215 traditional Chinese medicines are used in the prescription of traditional Chinese medicine,and the top ten drugs in the frequency of use are:Ligustrum lucidum,Eclipta chinensis,Cuscuta chinensis,Fructus gardeniae,Radix Bupleuri,Radix Paeoniae Alba,Rhizoma Chuanxiong,Angelica sinensis,Cortex Moutan,and Curcuma.The four gases are mainly cold and warm;The five flavors are mainly sweet,bitter and bitter;Meridians mainly belong to liver,kidney and heart channels.Cuscuta chinensis-Leonurus heterophyllus,Ligustrum lucidum-Echinacea,Bupleurum chinense-Echinacea-Ligustrum lucidum,Paeonia lactiflora-EchinaceaLigustrum lucidum alcohol are more commonly used drug compatibility.Through cluster analysis,it was found that the root of Cynanchum,roasted epimedium,coptis chinensis,scorched gardenia,peony peel,calcined oyster,ligusticum chuanxiong,rehmannia glutinosa,vinegar turtle shell,achyranthes bidentata;Radix Bupleuri,Radix Paeoniae Alba,Curcuma,Rhizoma Cyperi,Rhizoma Atractylodis with Bran,Fushen,Angelica sinensis;Vinegar Schisandra chinensis,lily,calcined magnetite,cauliflower,fried sour jujube kernel;Four cluster combinations of red peony,safflower,wine ligustrum lucidum,Eclipta chinensis,salt dodder,Chinese date,hypericum perforatum;Through complex network analysis,the core prescription drugs were determined to be Ligustrum lucidum,Gardenia jasminoides,Eclipta chinensis,Cuscuta chinensis,Bupleurum chinense,Ligusticum chuanxiong Hort,and Paeonia lactiflora.Conclusion:Professor Li Qiuyan’s core prescription for the treatment of climacteric syndrome consists of seven drugs,including Ligustrum lucidum,Gardenia jasminoides,Eclipta,Cuscuta chinensis,Bupleurum chinense,Chuanxiong,and Paeonia lactiflora.Professor Li Qiuyan believes that the pathogenesis of climacteric syndrome is based on kidney deficiency,liver depression,heat and blood stasis;The disease is located in the kidney,which is closely related to the liver and heart;The clinical treatment mainly focuses on tonifying the kidney,taking into account the treatment methods of soothing the liver and clearing the heart.Study 2:Based on network pharmacology and molecular docking,study the core prescription mechanism of Professor Li Qiuyan in the treatment of climacteric syndromeObjective:To explore the potential mechanism of the core prescription in the treatment of climacteric syndrome by analyzing and studying the active ingredients,core targets and action pathways of the core prescription in the treatment of climacteric syndrome through the network pharmacology method.Methods:Using the TCMSP database to search the active ingredients of Ligustrum lucidum,Eclipta chinensis,Gardenia jasminoides Ellis,Cuscuta chinensis,Bupleurum chinense,Ligusticum chuanxiong,and Paeonia lactiflora.The oral bioavailability≥ 30%and the drug-like property≥ 0.18 were used to screen,collect the active components and action targets,and use the UniProt database to standardize the names of the collected targets.Take "Climatic Syndrome" as the search term,search OMIM,TTD and GeneCards databases,and integrate the retrieved targets to obtain disease targets.Match the core prescription drug target with the disease target to obtain the intersection target.Use Cytoscape software to draw the component-target network diagram of the active ingredients of core prescription drugs in the remedy of climacteric syndrome,and screen the active ingredients;Through STRING database,protein interaction analysis was performed on the intersection target to screen the core target;Use Metascape database to conduct GO and KEGG enrichment analysis on intersection targets;The CB-Dock platform is used for molecular docking analysis of key components and core targets.Results:A total of 63 active ingredients and 197 action targets were screened from the core prescription.There are 4135 disease targets of climacteric syndrome.There are 149 core prescription and disease intersection targets.The active ingredients of the core prescription for the treatment of climacteric syndrome are quercetin,kaempferol,luteolin,isorhamnetin,catechin β-The core targets of sitosterol and stigmasterol are AKT1,TP53,IL6,TNF,VEGFA,ESR1,etc.,which are closely related to estrogen signal pathway,relaxin signal pathway,hypoxia-inducible factor-1 signal pathway,and apoptosis pathway.Molecular docking results showed that the key active ingredients had strong binding force with ESR1 and VEGFA.Conclusion:Professor Li Qiuyan’s core prescription for the treatment of climacteric syndrome may achieve the purpose of treating climacteric syndrome through antiinflammatory,antioxidant,anti-tumor and metabolic regulation.
