Objective: To mine the data of modern literatures on the treatment of sores and ulcers with encircling drugs in recent 38 years,explore the regularity of its prescription,carry out the network pharmacology analysis of the Chinese medicine combination with the highest support,investigate its mechanism in the treatment of venous ulcer,which is one of the sore diseases,so as to provide more basis for the treatment of sore with encircling drugs and guide clinical medication.Methods: This study was divided into two parts: Part Ⅰ was involved in studies on medication regularity of encircling drugs in the treatment of sore and ulcer based on the data mining technology.The digital journals in the Chinese Journal Fulltext Database(CJFD)and the modern literatures about encircling drugs in the treatment of sore and ulcer from 1984 to 2022 in the database of VIP China Science and Technology Journal Database were searched.The prescription database of hoop drugs for treating sores was established by Excel table,and statistical analysis was carried out.The association rules analysis and cluster analysis,principal component analysis were carried out by IBM SPSS Modeler and IBM SPSS Statistics 26.0,respectively.Part Ⅱ was involved in exploration on the mechanism of the drug group with the highest support in the treatment of venous ulcer by using network pharmacology.Using TCMSP database to search the active ingredients of the drug group with the highest support and their corresponding prediction targets,the target information was corrected in Uniprot database,the related targets of venous ulcer were retrieved by Gene Cards database,the component-target network diagram of the drug group with the highest support was constructed by Cytoscape3.7.2,the protein interaction analysis was carried out in STRING platform,and the GO function and KEGG enrichment analysis were carried out in Metascape database.Result:1.This study included 400 modern literatures on encircling drugs in the treatment of sores and ulcers,involving 402 prescriptions and 180 traditional Chinese medicines.The top 10 traditional Chinese medicines used most frequently were Rheum officinale,Cortex phellodendri,borneol,Angelica dahurica,realgar,Atractylodes lancea,Radix Trichosanthis,licorice,Indigo naturalis and turmeric in turn;the top four traditional Chinese medicines used frequently are as follows: heat-clearing drugs,blood circulation-promoting and blood stasis-removing drugs,purgative drugs,toxin-attacking insecticidal and antipruritic drugs.2.A total of 44 strongly correlated drug combinations were obtained by association rule analysis,among which the drug group with the highest support was "Cortex Phellodendri→Angelica dahurica";Cluster analysis shows that there are 5 cluster groups which are more in line with the actual situation.7 common factors were extracted by principal component analysis,and the highest contribution factors were Curcuma longa,Tangerine Peel,Rhizoma arisaematis,Rhizoma Atractylodis,Magnolia officinalis,Trichosanthin,Angelica dahurica,Glycyrrhiza uralensis Fisch,Phellodendron amurense and Rheum officinale.3.Through network pharmacological research,342 active ingredients and 148 predicted targets of the drug group "Cortex Phellodendri→Angelica dahurica" with the highest support were collected,187 targets of venous ulcer disease were extracted,62 common targets of active ingredients and diseases were obtained,31 biological processes,5 cellular components,8 molecular functions and 13 significant signal pathways were obtained by enrichment analysis.Conclusions: 1.The main treatment principles of treating sores and ulcers with hoop-around drugs are clearing heat and purging fire,cooling blood and detoxifying,promoting blood circulation and removing blood stasis,eliminating dampness and relieving itching.According to pathogenesis,it is assisted by eliminating dampness and promoting qi,inducing resuscitation and awakening mind,enriching blood and qi,eliminating phlegm and resolving stagnation,and dispersing exterior evil.2.The main active components of "Cortex Phellodendri→Angelica dahurica" in the treatment of venous ulcer include quercetin,β-sitosterol,tetrahydroberberine,phellodendrine,dauricine,etc.It has anti-inflammatory,antibacterial,antioxidant,reducing inflammatory injury,improving vascular endothelium,promoting wound healing,etc."Cortex Phellodendri→Angelica dahurica" has played a therapeutic role on venous ulcer through VEGFA,IL-1β,IL-6,TNF and other targets,hypoxia inducible factor 1 signaling pathway,nuclear factor-κB signaling pathway,cancer signaling pathway and many other pathways. |
