Dermatological Toxicology And Pharmacodynamics Of Qibaomei Beard Tincture

Objective:in this study,through animal experiments,to explore the acute and chronic toxicity of Qibao Meike beard on the skin of SD rats,irritant experiments on white rabbits and allergic experiments on guinea pigs,to explore the skin toxicology of Qibao Meike beard tincture in the treatment of alopecia areata,so as to help better use Qibao Meike beard tincture in clinical practice.Objective to study the effect of Qibao Meike tincture on the growth cycle of hair and hair follicle in SD rats.In order to better use Qibao Meike tincture in clinical.Methods:Qibao Meike beard prescription and 70% ethanol solution were prepared into four different concentrations of Qibao Meike beard tincture solution:100%、70%、50% and 30%.According to the random principle,the70% Qibao Meike beard tincture of SD rats acute toxicity experiment,100%,70%,50%,30% Qibao Meike beard tincture of SD rats chronic toxicity experiments,will be 70%,30% Qibao Meike beards tincture effect in the back of the white rabbit skin irritation experiments,and 30%Qibao Meike beards tincture effect on guinea pig skin allergic skin line the sensitivity test.In cutaneous pharmacodynamics study,SD rats were randomly divided into 6 groups(n=10)according to the randomization principle:10% Qibao Meike tincture group,30% Qibao Meike tincture group,50% Qibao Meike tincture group,70% Qibao Meike tincture group,Minodil group,blank control group,external rub twice a day,each time1 ml,after medication in local massage 3-5 circles to drug absorption,daily observation of skin and hair,with dermoscope to collect images,observation period 21 d,and according to the corresponding indexes such as area,new length,dermoscopic and histopathological sections were observed to evaluate the efficacy.Results:in SD rat skin acute toxicity test,skin to observe the clinica lchanges of rats after medicine,spirit,activity,diet,sleep,mainland,breath,eyes,mucous membrane,fur,no obvious change,during the experiments,groups of animals did not see death,before the trial,7 days after delivery respectively record each animal body weight change,weight no significant differences between groups(P>0.05).In SD rat skin chronic toxicity test,four groups of animals after test,the colour of rats,independent activity,breathing,eating,spirit,sleep,mainland,eyes,mucous membrane,no obvious change,during the experiments,groups of animals did not see death,the last 24 h after the treatment,each SD rats,hepatic and renal function of hematology indexes comparison,there were no statistically significant difference(P>0.05).The results of skin histological examination showed that the results of epidermis,dermis and subcutaneous tissue were intact,without structural disorder,bleeding spots,edema,epidermis thickening,degeneration,necrosis,and no significant difference between the administration area and the control area.In the irritation experiment of white rabbits,no obvious erythema and edema were observed,the skin was smooth,and normal new hair could be seen in all groups after 7 days of continuous administration and 7 days after withdrawal.The skin wounds on both sides of the back of each damaged skin group healed generally.However,70% Qibao Meike tincture,95% ethanol intact skin group and 70% Qibao Meike tincture,95%ethanol damaged skin group each had 1 skin yellow staining,95% ethanol intact skin group had 1 hair adhesion,yellow,shaving device could not shave normally.After cleaning the drug,there was no obvious change in the yellow stained skin,the adhesive hair was loosened,and the skin color was normal.In Qibao Meike beards tincture of guinea pig skin allergic experiment process,drug and excipients group guinea pigs skin did not appears erythema,edama,two groups of animals did not appear asthma,dyspnea,unstable,shock,systemic allergic reaction,such as positive control group excitation visible after 9,occurrence erythema of floor of guinea pigs and mild edema,nine erythema significantly increased after 24 hours,48 hours slightly faded red spot.The erythema gradually subsided within 72 hours.During the experiment,no systemic allergic symptoms such as asthma,dyspnea,unstable standing and shock were observed in the positive control group,and no animal died.In the pharmacodynamic experiment,the change time of skin from red to white was significantly reduced in Qibao Meike tincture group(P<0.05).The hair length of the tincture group was longer than that of the control group at 21d(P<0.05),and there was no significant difference between the 50% Qibao Meike tincture group and the minoxidil group(P>0.05).Compared with blank control group,the number of hair folicles in 50% and 70% Qibao Meike tincture groups was increased(P<0.05).Compared with Minoxidil tincture group,the number of folicles in 50% Qibao Meike tincture group was smaller(P>0.05).Compared with blank control group,the thickness of the dermis was increased in 30% Qibao Meike tincture group,50% Qibao Meike tincture group and 70% Qibao Meike tincture group(P<0.05).Conclusion:Qibao Meike tincture anhydrin as a topical drug with high safety and less adverse reactions,has a good application prospect.In the skin pharmacodynamic experiment,it was found that Qibao Meike tincture can accelerate the hair growth of rats and make the hair follicle growth period of rats as soon as possible.The efficacy of 50% concentration is close to Minoxidil tincture,and no obvious adverse reactions were observed.
