Objective:The aging model of HaCaT cells was established by UVB irradiation.The effect of UVB on HaCaT cell senescence model was observed by using Saussurea medusa Maxim polysaccharide.The changes of key molecular levels of ERK1/2 and AQP-3 were measured to elucidate the molecular mechanism of saussa medusa polysaccharide in reducing skin photoaging and maintaining skin barrier to elucidate the molecular mechanism of Saussurea medusa Maxim polysaccharide alleviating skin photoaging.Methods:HaCaT cells were cultured by in vitro culture technique,and the target cells were inoculated into a 24-well plate containing slime,and randomly divided into four groups with 3 Wells in each group: Normal group,UVB group,low-SMMP group and high-SMMP group.The cell aging model was established by UVB radiation for 1h.The drug group was treated with Saussurea medusa Maxim polysaccharide,and then the four groups were cultured for 18 h.Annexin V-FITC/PI kit was used to detect apoptosis and necrosis of cells in each group.The effects of Saussurea medusa Maxim polysaccharide on the expression levels of ERK1/2 and AQP-3 in photoaged HaCaT cells were detected by immunohistochemistry.Results:Annexin V-FITC/PI kit for cell apoptosis and necrosis detection showed that compared with Normal group,cell apoptosis and necrosis in UVB group were significantly increased(P<0.05),suggesting that UVB radiation can accelerate cell apoptosis and induce cell necrosis,indicating that cell senescence model was successfully established.The apoptosis of low-SMMP group was decreased compared with UVB group(P<0.05),and the apoptosis of cells in the high-SMMP group was significantly decreased compared with that in the UVB group(P<0.05),indicating that the Saussurea medusa Maxim polysaccharide had significant significance in reducing cell apoptosis and necrosis.Immunohistochemical results showed that the expression rate of ERK1/2 positive cells in UVB group was higher than that in Normal group(P <0.05).The expression rate of ERK1/2 positive cells in low-SMMP group was lower than that in UVB group(P <0.05).The expression rate of ERK1/2positive cells in high-SMMP group was lower than that in UVB group(P<0.05).The expression rate of ERK1/2 in high-SMMP group was lower than that in low-SMMP group(P<0.05).The expression rate of AQP-3 in UVB group was lower than that in Normal group(P<0.05).The expression rate of AQP-3 positive cells in low-SMMP group was higher than that in UVB group(P <0.05).The expression rate of AQP-3positive cells in high-SMMP group was higher than that in UVB group(P<0.05).The expression rate of AQP-3 positive cells in high-SMMP group was higher than that in low-SMMP group(P<0.05).Conclusion:Saussurea medusa Maxim polysaccharide has a certain effect on the repair of UVB-induced photoaging HaCaT cells. |
