Objective: In this study,Saussurea medusa maxim polysaccharide was used as the research object to observe the effects of Saussurea medusa polysaccharide on skin appearance changes,skin water content and AQP-3 expression in mouse skin aging models,and explore the possible anti-aging mechanism of Saussurea medusa polysaccharide.Methods: A total of 75 healthy SPF-grade KM male mice were selected and randomly divided into 5 groups,each with 15 mice;normal group,aging model group,VE group,low-SMMP group,and high-SMMP.The aging skin model of mice was established by D-galactose combined with UVB radiation,and the meds such as Saussurea medusa polysaccharide and vitamin E were orally intervened.During the modeling of mice,the mice in each group were photographed,and the skin color,wrinkles,smoothness,and erythema desquamation and other appearance characteristics and general conditions of the mice’s skin were roughly observed.The effects of Polysaccharide from Saussurea medusa on water content and AQP-3 expression in aging skin of mice were observed by oven drying and immunohistochemistry.Results:1.The skin changes of the mice were observed by naked eyes.Compared with the Normal group,the skin of the UVB group gradually became rough,dry,lacking luster,and the skin appearance was leather-like,with static deep wrinkles,a large number of erythema and desquamation,and poor elasticity.After the intervention of drug gavage,the skin of mice in each group showed obvious improvement in different degrees,their wrinkles became thinner and shallower,desquamation was reduced,the skin was smoother,and the elasticity was better.Among them,the skin changes in the HighSMMP group were particularly close to those in the Normal group;compared with the Normal group,the UVB group had significantly lower skin water content(P<0.05);compared with the UVB group,the VE group,Low-SMMP group,and High-SMMP group The skin water content of the group increased(P<0.05),especially the effect of High-SMMP was the most significant.2.Compared with normal group,the expression of AQP-3 in UVB group was significantly lower(P < 0.05);compared with UVB group,the expression of AQP3 protein in VE group and each Saussurea medusa group was increased in varying degrees(P < 0.05),and the effect of high SMMP group was better than that of VE group and low SMMP group.Conclusion: Saussurea medusa polysaccharide can protect the aging skin of mice to a certain extent by increasing the water content of aging skin and the expression of AQP-3,so as to achieve the purpose of delaying aging.
