objective: To investigate the effects of Saussurea medusa Maxim on the expression of extracellular regulated protein kinase(ERK)and matrix metalloproteinase(MMP)-1 in photoaged human keratinocytes(Ha Ca T)and the protective effect of Saussurea Medusa Maxim on photoaged Ha Ca T cells.Methods: Ha Ca T cells were cultured in a cell incubator with 5% carbon dioxide at 37℃.The Ha Ca T cells were used for subculture when the degree of growth and fusion of Ha Ca T cells reached 80-90% under the microscope.The Ha Ca T cells used in this experiment were 3 to 5 generations in logarithmic growth period.After Ha Ca T cells were cultured,Ha Ca T cells were divided into normal group(containing only Ha Ca T cells,without any intervention),UVB irradiation group(UVB irradiation only,30 m J/cm~2),UVB irradiation + low-dose SAussa medusa polysaccharide group(30m J/cm~2+10mg/m L),UVB irradiation + high-dose Saussa medusa polysaccharide group(30m J/cm~2+40mg/m L).After the Ha Ca T cell intervention was completed,the expression levels of ERK and MMP-1 proteins in each group were detected by immunohistochemistry,and the proliferation activity of Ha Ca T cells was detected by CCK-8 method.Results: Compared with UVB group,the expression levels of ERK1/2 and MMP-1 proteins in Ha Ca T cells of Saussurea medusa Maxim(low and high dose)groups showed a downward trend;Compared with UVB group,the proliferation rate of Ha Ca T cells in Saussurea medusa Maxim(low and high dose)groups increased.Conclusion: Saussurea medusa polysaccharide can protect Ha Ca T cells after UVB radiation. |
