[Objective]: Atractylodes lancea is a kind of traditional Chinese medicine which is commonly used in clinic.The "frost" is a one of the important quality evaluation characteristics of A.lancea.However,the cause of “frost” phenomenon has not been scientifically revealed.Therefore,this Primary aims to solve the following problems.(1)We summarized Herbal textual research of “frost” phenomenon of A.lancea,to research provide herbalism basis for the modern study of “frost” medicinal materials.(2)The “forst” characteristics and chemical components of A.lancea from different regions were compared;(3)Comparied the volatile oil components of A.lancea from different regions;(4)Clarified the distribution of “frost” and volatile oil in the rhizomes of A.lancea.[Method]: This paper made textryal research and sorted out the historical origin of“frost” of A.lancea;The rhizome section and “frost” of A.lancea collected from three regions were observed with stereomicroscope and metalloscope when they were fresh,and the components of “frost” were determined by GC-MS;The research group analyzed the volatile oil components in different organs of A.lancea from four regions by using GC-MS,and quatitative analysis of rhizome was carried out to clarify the content difference of volatile oil in A.lancea from different regions;Laser microdissection combined with GC-MS was used to construct the correlation between different organs and chemical components of A.lancea,and to clarify the accumulation site of "frost" of A.lancea.[Result]:(1)In the Ben Cao Yao Pin Shi Di Zhi Guan Cha(1937),it was recorded that A.lancea was “placed for 1-2 days,the white and clean powder and frost were precipitated out”.“Frost” has gradually become one of the quality evaluation characteristics of A lancea,and it together with "Cinnabar Point" has become an important distinguishing point between A.lancea and A.Chinensis.(2)Through the observation of the rhizomes of two chemical types,it was found that there were many oil chambers scattered on the cross section of two chemical types of A.lancea,and white fine needle crystals could be precipitated at it too.But the “frost” phenomenon of the section of Jiangsu Maoshan type of A.lancea was not obvious,while the "frost" phenomenon of Dabieshan type of A.lancea was more obvious.The "frost" of A.lancea from three regions were analyzed by GC-MS,and the results showed that the "frost" was composed of hinesol and β-eudesmol.(3)The peak area normalization method was used to determine the relative contents of the chemical components in two chemical types of A.lancea.The results showed that 21 components were identified in A.lancea from different regions,and the rhizome accounted for the most chemical components,followed by fibrous root,flower,leaf and stem.For rhizomes from Henan Province,Anhui Province,Hubei Province,Jiangsu Province,the ratios of hinesol,β-eudesmol,atractylon and atractylodin were(12.66:11.71:0.99:1),(17.06:4.55:0:1),(7.43:6.23:0:1)and(0.13:0.16:1.52:1)respectively.(4)The different tissue parts of the rhizome of A.lancea from four regions were identified by Laser Microdissection.The cutting parts were periderm,cortex,oil chamber in periderm,secondary phloem,oil chamber in secondary phloem,secondary xylem,oil chamber in secondary xylem,vascular ray,pith and oil chamber in pith.The results showed that the volatile oil of the A.lancea rhizome was mainly distributed in the perderm and oil chamber,and five components(hinesol,β-eudesmol,atractylon and atractylodin)were identified in total,but no volatile oil was detected in cortex,secondary phloem,secondary xylem,vascular ray and pith.The types and contents of volatile oil in rhizome have great differences.The main components of A.lancea in Dabieshan type were hinesol,β-eudesmol and elemol.Among them,hinesol and β-eudesmol(“frost” volatile oil)from Anhui and Henan are mainly distributed in oil chambers of 3 parts(oil chamber in coxter,oil chamber in secondary xylem and oil chamber in pith),and the “frost”volatile oil of A.lancea in Hubei Provinve was mainly distributed in the oil chamber in secondary phloem,oil chamber in secondary xylem and pith in pith.In Maoshan type,the mian components were atractylon,atractylodin,hinesol and β-eudesmol,but the relative content of “frost” volatile oil was low,which were mainly distributed in oil chamber in cortex and oil chamber in secondary phloem.[Conclusion]:(1)“Frost” is one of the important characters reflecting the excellent quality of A.lancea.(2)The “frost” of A.lacnea was white fine needle;the "frost" of different chemical types of A.lancea had obvious diffenence,that was,the Jiangsu Maoshan type of A.lancea was almost no "frost" phenomenon,and the "frost" phenomenon of Dabieshan type of A.lancea was obvious;The "frost" components of different chemical types of A.lancea were hinesol and β-eudesmol.(3)There were obvious differences in the volatile oil components of different chemical types of A.lancea,atractylon and atractylodin were the main components of the volatile oil in Maoshan type of A.lancea,and hinesol and β-eudesmol were the main components of the volatile oil in Dabieshan type of A.lancea;the ratios of volatile oil components in rhizomes of A.lancea with different chemical types were verified.(4)The relative content of volatile oil of hinesol and β-eudesmol in the oil chamber of A.lancea was the highest,and the content of volatile oil in the periderm and other parts was low or not.Therefore,it was speculated that hinesol and β-eudesmol are mainly accumulated in the oil chamber,and they were separated from the cross section of A.lancea through the oil chamber,forming a special "frost" phenomenon of A.lancea. |
