This thesis was performed for quality control of Atractylodes lancea(Thunb.) DC..In the whole reaserch,"famous-region drugs" from Maoshan region was used to be the benchmark.Its characteristic components was studied and compared to the drugs from other different places by modern analysis methods.Theses studies were performed on the basis of the clinical effects of Atractylodes lancea(Thunb.) DC.. The fingerprint was also founded to supply the evidence for evalutating the quality of Atractylodes lancea(Thunb.) DC..The main contents are as following:1.The volatile components of Atractylodes lancea were pured by column chromatography and TLC.Six main compounds(e.g.atractylone,hinesol,β-eudesmol, atrctylodin,atractylenolideⅡand acetylatractylodinol) were isolated and identified by studying their physical and chemical properties and spectrum.The purity of theses compounds were analyzed,and the result showed their purity were higher than 98%.2.A GC method for simultaneous determination of 4 compounds(atractylone, hinesol,β-eudesmol and atrctylodin) in Atractylodes lancea(Thunb.)DC,was established.The result of methodological study showed that this method was simple, specific,repeatable and stable.It can be applied for the simultaneous determination of 4 compounds(atractylone,hinesol,β-eudesmol and atrctylodin) in Atractylodes lancea(Thunb.) DC..The contents of this four compounds in 15 samples from different places were determined,and the result showed that the contents of these four compounds were significantly different between samples from famous-region and unfamous-region.The result of clustering analysis by SPSS software showed that 15 samples were divided into two main clusters.Samples from famous-region and samples from Nanjing Lingshan region and Yixing area of Jiangsu province and samples from Yingshan region of Hubei province were one cluster.Another cluster included samples from Huoshan region of Anhui province and samples from Henan and Shaanxi province.3.A convenient and sensitive GC-MS method was developed to identify 13 sesquiterpenes and polyacetylenes(e.g.caryophyllene,γ-elemene,α-caryophyllene, β-selinene,isoledene,elixene,β-Vatirenene,atractylone,hinesol,β-eudesmol, atrctylodin,atractylenolideⅡand acetylatractylodinol) in Atractylodes lancea.Among those compounds,four major components including atractylone,hinesol,β-eudesmol and atrctylodin were quantified with standards;contents of other components were estimated based on compounds with similar structures.In this study,we presented that the concentrations of those 13 components varied drastically in Atractylodes lancea samples from different producing areas.Among those components,atractylenolideⅡand acetylatractylodinol were identified by GC-MS for the first time.A hierarchical clustering analysis based on relative peak areas of those 13 components in total ion current(TIC) profiles was used to characterize Atractylodes lancea samples from different producing areas.Further clustering analysis showed that a simplified method with only four major bioactive components could be used to serve the same aim. Sample from Nanshan region of Jiangsu province,as well as those samples from Yingshan region of Hubei province and Bozhou area of Anhui province showed the similar chemical compositions with those famous region drugs.That group of samples was characterized with having atractylone,hinesol,β-eudesmol and atrctylodin as their main components.Sample from Yixing areas of Jiangsu province,as well as sample from Jimingshan and Luotian area of Hubei province showed the similar chemical compositions with those samples from Henan and Shaanxi province.That group was characterized with only containing hinesol andβ-eudesmol as major components.4.The fingerprint of three parts with different polarity of water extract of Rhizoma Atractylodis was studied.The result showed that there were 10 main common peaks in petroleum ether extract,11 main common peaks in Ethyl acetate extract,and 14 main common peaks in butanol extact.The characers of samples were more comprehensively reflected by the informination of common peaks in the three different extracts.The result of similarity calculation about these three extracts showed that samples from famous-region were very similar,and the order of similarity between samples from famous-region and unfamous-region was Nanjing Lingshan region of Jiangsu provience,Anhui provience,Hubei provience,Henan provience and Shaanxi provience.The result of clustering analysis with the three different extracts together by SPSS software showed that 16 samples were divided into two main clusters.Samples from famous-region and samples from Nanjing Lingshan region,samples from Anhui province and Hubei province were one cluster. Another cluster included samples from Henan and Shaanxi province.This clustering analysis result was similar with the result studied in the volatile fraction.Our study showed that it was more reasonable to evaluate the qaulity of Atractylodes lancea with the comprehensive characteristics of volatile and water soluble components.
